| Objective:To investigate the effects of different sites of Tuina on the behavior and intestinal flora of autistic spectrum disorder(ASD)model rats with valproic acid(VPA)-induced,and explore the relationship between Tuina,intestinal flora,and autism-like behaviors.Methods:VPA(600 mg/kg)and equal amounts of saline were inj ected intraperitoneally into 12.5D Sprague Dawley rats at gestation,respectively,and the male offspring of saline group were the control group,and the male off spring of valproic acid group were randomly divided into 3 groups:model group,dorsal group,abdominal group after model validation(each n=7).Growth and development tests were performed at D7,D14,and D21 after birth,behavior of the four groups was tested at D21,and the Tuina intervention using the selfmade push stimulation was started twice a day for 14 days for the dorsal and abdominal groups,a total of 28 times.Behavior and intestinal contents were tested at the end of the intervention at D35D38.Results:VPA and saline rats showed significant differences in weight,length length,tail length and eye opening(P<0.01),which verified the reliability of this experimental model.At the behavioral level,the social new rat time and central area movement time was increased(P<0.05),the number of buried was reduced(P<0.01)in the dorsal group compared with the model group on day D35,their social interactions,anxiety,and stereotypical behaviors have been improved.And the number of buried was reduced(P<0.05)in the abdominal group,stereotypic behavior improvement.Compared with the abdominal group after intervention,the results of the dorsal group were not significantly different in the three-chamber social and open field tests(P>0.05).The dorsal group showed reduced number of buried beads and more significant improvement in stereotypic behaviors compared with the abdominal group on D35(P<0.05).On the level of intestinal microorganisms:The results of the diversity analysis show that the flora richness increased(P<0.05)and the diversity increased in both dorsal and abdominal groups,with P<0.05 in the dorsal group and P<0.01 in the abdominal group.The results of differential analysis indicated that at the phylum level,compared with the model group,the relative abundance of Firmicutes in the abdominal group showed a significant reduction trend(P<0.05);At the genus level,the relative abundance of Lactobacillus in the dorsal group and abdominal group decreased significantly compared with the model group(P<0.05).The marker species Blautia in the dorsal group increased significantly(P<0.05).Conclusion:Tuina can improve the behavior and intestinal flora structure of VPA-induced ASD model rats,increase microbial richness and diversity in the gut,more significant improvement in ASD behavior by dorsal intervention and may be related to an increase of Blautia in the gut. |
