Objective:Based on the theory of supramolecular chemistry,the change of the guest "imprinted template" during the processing of Rhizoma Polygonati was studied,and the quality inspection results were combined to lay a foundation for the quality research of Rhizoma Polygonati.Method:(1)Self made raw materials and slices of Rhizoma Polygonati prepared by steaming and baking until twelve steaming and twelve baking were determined.The inspection items,extracts,and polysaccharide content specified in the 2020 edition of the Pharmacopoeia were determined,and characteristic components such as chromaticity and 5hydroxymethylfurfural were added to the reference literature for content determination.(2)Establish a multi steam Rhizoma Polygonati UPLC fingerprint from the aspects of extraction method,detection wavelength,and selection of mobile phase,and compare the characteristics of changes.(3)Starting from the UPLC fingerprint results using supramolecular theory,compare the total statistical moment parameters and total statistical moment similarity of Rhizoma Polygonati with different steaming times;GC-MS analysis was carried out on Rhizoma Polygonati Rhizoma prepared with different times of steaming.The principle of total statistical moment and the principle of information entropy were used to compare the difference of total statistical moment parameters,calculate the molecular connectivity index(MCI)of the contained ingredients,and study the change rule of "imprinting template" between the decoction pieces.Result:(1)With the increase of steaming times,the L*and E*ab values of Rhizoma Polygonati powder first showed a downward trend,and remained basically stable after four steaming,indicating that the brightness of Rhizoma Polygonati gradually decreased,manifested as bright to dark;The values of a*and b*show a trend of first increasing,then decreasing,and finally stabilizing,indicating that the color of Rhizoma Polygonati powder transitions from light yellow to reddish brown one by one,followed by a gradual decay of red and yellow,transforming into black brown to lacquer black.The indicators such as leaching rate,polysaccharides,total saponins,and 5hydroxymethylfurfural show different changes with the increase of steaming times.The overall leaching rate shows a trend of first increasing and then decreasing,the overall polysaccharide content shows a trend of decreasing and finally stabilizing,and the total saponin content shows a gradual increase and finally stabilizing.The content of 5hydroxymethylfurfural in the raw product is 0,and its content increases with the increase of steaming times.(2)The chromatographic conditions of Rhizoma Polygonati UPLC fingerprint are as follows:the mobile phase is acetonitrile A-aqueous solution B(0-3 minutes,1%A;3-18 minutes,1%-30%A;18-27 minutes,30%-100%A;27-30 minutes,100%-1%A);Column temperature 30℃;The flow rate is 0.3 mL/min;Inj ection volume is 2 μL;The detection wavelength is 200nm;(3)In terms of the total statistical moment parameters of Rhizoma Polygonati UPLC,the RSD values of AUCT,MCRT and VCRT were 25.87%,7.74%and 29.43%respectively after the second steaming.From the third steaming to the twelfth steaming,the similarity of the total statistical moment reached more than 0.85,and the maximum information entropy reached 4.83 after the fifth steaming;In terms of GC total statistical moment parameters,after the first steaming,the RSD values of AUCT,MCRT,and VCRT were 3 4.31%,5.10%,and 31.67%respectively.After three steaming and three drying,to twelve steaming and twelve drying,the similarity between the pieces basically reached more than 0.85,and after four steaming,the information entropy reached the maximum value of 3.74;There are 17 characteristic "imprinting templates"(also known as material foundations),and as the MCI order of the pathway increases,the number of missing cases increases;The higher the order of MCI in the ring,the fewer missing cases.The distribution of MCI at various levels is both concentrated and discrete,with the majority being concentrated overall.Conclusion:There are 17 characteristic "imprinting templates"(also known as material basis)of Polygonatum,and their number decreases with the increase of steaming times.Chain structures are mostly small molecule compounds,while ring structures are mostly large molecule compounds.During the processing of Rhizoma Polygonati,changes in supramolecular chemical parameters,combined with changes in chromaticity,determination of extract content,and effective component content,suggest that steaming Rhizoma Polygonati 3 to 5 times is more effective in terms of quality.
