OBJECTIVE: To explore the pattern of drug use in Chinese medicine for primary hyperhidrosis through data mining,and to explore the mechanism of action of the most supported drugs with the help of network pharmacology,so as to provide theoretical basis and guiding ideas for clinical treatment of primary hyperhidrosis.Data and methods:(1)We searched the modern literature of clinical research and clinical observation on primary hyperhidrosis in China Knowledge Network,Wanfang and Vipul databases,and collected prescriptions of Chinese medicine.The database was established based on Microsoft Excel 2019,and pivot tables were applied to obtain highfrequency drugs;association rule analysis was performed based on IBM SPSS Modeler18.0 to obtain core drugs;cluster analysis and prediction were performed based on IBM SPSS Statistics 25.0 to mine the medication pattern of TCM for primary hyperhidrosis.(2)To obtain the active ingredients and action targets of the most supported drugs through TCMSP database;to obtain disease targets based on Pharm GK database and Gene Cards database;to obtain intersection genes using Venn online tool;to construct TCMcomponent-target network diagram using Cytoscape3.7.2 software;to construct TCMtarget network diagram based on String database and Cytoscape3.7.2 software was used to construct the PPI network and core targets;the Metascape database was used to perform GO functional enrichment analysis and KEGG pathway analysis on the potential intersection target genes to obtain the most supported potential mechanism of action of drugs for primary hyperhidrosis.Results:1.By frequency analysis of 107 Chinese herbal medicines in 115 prescriptions(total frequency 1048 times),the high frequency drugs(>50)were: scutellaria baicalensis,oyster,roasted licorice,floating wheat and bryophyllum.2.Distribution of efficacy,four qi,five tastes and meridians: the top 3 used in the category of efficacy(class II classification)in terms of frequency: qi tonic,fixing the surface to stop sweating,and blood tonic;the top 3 in the distribution of the four qi of drugs: warm,flat and slightly warm;the top 3 in the distribution of the five tastes of drugs:sweet,bitter and sour(astringent);the distribution of the meridians of drugs: the heart meridian was the most frequent,followed by the spleen and liver meridians.3.The results of association rule analysis showed that the pair with the highest support of association rule was "Astragalus-Bryophyllum";the pair with the highest number of links was "Astragalus-Oyster";the pair with the highest support of third-order association rule was "Oyster-Oyster-Astragalus".The drug pair with the highest support of the third-order association rule was "Oyster-Astragalus".There were 125 groups of drug combinations with confidence level 1.4.Cluster analysis was performed to obtain three groups of prescriptions:(1)Atractylodes macrocephala,Anemarrhena,Astragalus,Wu Wei Zi;(2)Oyster,Dragon Bone,Roasted Licorice,White Peony;(3)Radix Angelicae Sinensis,Radix et Rhizoma,Fructus Wheat,Ephedra root.5.The top five active ingredients of Astragalus-Brassicae are quercetin,kaempferol,7-Omethyl-isoconvex sarcosol,mangosteen,isorhamnetin;among the top ten active ingredients,only acetoxycanthone is from Astragalus,and the remaining nine active ingredients are from Astragalus;the core targets include: MMP9,CXCL8,JUN,IL CXCL8,JUN,IL10,AKT1,IL6,CASP3,IL1 B,ICAM1,HIF1 A,MMP2,VEGFA,EGF,CCL2,IL4;mainly by acting on JAK-STAT signaling pathway,AGE-RAGE signaling pathway,VEGF signaling pathway,VEGF signaling pathway and VEGF signaling pathway.VEGF signaling pathway,P53 signaling pathway,Calcium signaling pathway,AMPK signaling pathway,etc.Conclusion:1.The high frequency drugs for the treatment of primary hyperhidrosis are radix astragali,oyster,roasted licorice,and floating wheat,Atractylodes macrocephala.2.In primary hyperhidrosis,"sweet and warm" products are used,with emphasis on "tonifying and astringent",and the drugs are mainly applied to the heart meridian.3.The drug pair with the highest support for the association rule is: "Huang Qi-Bai Zhu".4.In the treatment of primary hyperhidrosis,tonification is used to treat the symptoms,and astringency is used to treat the root;most of the formulas are formulated with "tonic" and "astringent" methods.5.The grouped formulas are Yu Ping Feng San,Gui Zhi plus Dragon Bone and Oyster Soup and Sheng Di Huang Decoction with reduction,which can be used as primary hyperhidrosis lung and spleen qi deficiency formula,yin and yang imbalance formula and yin deficiency fire formula respectively.6."Astragalus-Bryophyllum" is used to treat primary hyperhidrosis through multiple active ingredients,acting on multiple targets and regulating multiple pathways to exert antiinflammatory,antioxidant,immune regulating,vascular growth and cell proliferation,and neuroprotective effects.
