| Objective:Functional dyspepsia(FD)can be caused by abnormal gastric motility,Helicobacter pylori infection,intestinal flora disorder,anxiety,depression and other causes,among which the imbalance of brain-gut axis(Brain-gutaxis,BGA)occupies a key position.In this experiment,cholecystokinin(cholecystokinin,CCK)and leptin(Leptin,LP)were used as the foothold,and the compound model of "irregular feeding combined with tail clamping" was used to establish the model of FD rats.The traditional Chinese medicine compound Weixing granule combined with electroacupuncture at "Zusanli" and "Neiguan" points was used as the treatment.To observe its effects on hair texture,depression,anxiety,food and water intake,fecal texture,body weight,preference for sugar water,intestinal propulsion rate,content of LP and CCK in serum,expression of LP and CCK in stomach and brain tissue of FD rats,to explore the effect and related mechanism of traditional Chinese medicine compound prescription combined with electroacupuncture in the treatment of FD,so as to provide experimental basis for clinical promotion of acupuncture combined with medicine therapy.Materials and methods:A total of 50 healthy SPF male SD rats aged 8 weeks were selected.After 5 days of adaptive feeding,the rats were randomly divided into normal group,FD model group,electroacupuncture group,traditional Chinese medicine group and acupuncture group,with 10 rats in each group.Except for the normal group,the other four groups of rats were made FD rat model with the method of irregular feeding and tail clamping for 4 weeks.In the intervention stage,the rats in the traditional Chinese medicine group were treated with compound Weixing granule,the concentration of the solution was 0.08g/m L,and the intragastric dose was 10 m L/kg/ day.In the electroacupuncture group,the rats were first bound and fixed,and the bilateral "Zusanli" and "Neiguan" acupoints were directly punctured at 5mm and 2mm,respectively,connected with the electroacupuncture instrument,continuous wave,2Hz at 15 min / day.The rats in the acupuncture group were treated with traditional Chinese medicine intragastric administration and electroacupuncture in turn.The specific operation methods were the same as the traditional Chinese medicine group and the electroacupuncture group.The rats in the traditional Chinese medicine,electroacupuncture and acupuncture group were intervened continuously for 7 days,once a day.During the intervention period,rats in the normal group and FD model group were also stimulated by entrapment,once a day for 7 days.Observe the general condition,weight gain,sugar preference,gastric residual rate and intestinal propulsion rate of rats in each group;observe the pathomorphology of gastric antrum smooth muscle tissue by HE staining;detect the content of LP and CCK in serum by enzyme-linked immunosorbent assay(ELISA);detect the protein expression of LP and CCK in stomach and brain tissue by immunohistochemical method.Results:1.After modeling,except for the normal group,the other four groups had frequent emotions such as rough hair,yellow hair,tension and anxiety,like pricking and pushing,reduced activity,reduced food and water intake,soft and irregular stool texture,and so on.Compared with the normal group,the body weight of the rats in these four groups not only slowed down,but also the preference for sugar water decreased significantly(P < 0.05),the residual rate in stomach increased significantly,and the intestinal propulsion rate decreased significantly(P <0.05).2.After sampling,no abnormal conditions such as hemorrhage,edema,erosion and ulcer were found in gastrointestinal tissue of rats in each group under naked eye observation,and a small amount of neutrophils and eosinophils were found in gastric antrum tissue of rats in FD model group under microscope.3.After 7 days of intervention,the hair of the rats in the traditional Chinese medicine group,electroacupuncture group and acupuncture group became softer and brighter,the emotions such as irritability,anger and anxiety were relieved,the stool gradually took shape,and the intake of food and water increased significantly.Compared with the FD model group,the weight gain and sugar preference of rats in the traditional Chinese medicine group,electroacupuncture group and acupuncture group were significantly increased,especially in the acupuncture group,and the weight gain rate in the traditional Chinese medicine group was also significantly higher than that in the electroacupuncture group,but there was no significant difference between the electroacupuncture group and the electroacupuncture group.4.After intervention,compared with FD model group,the residual rate in stomach of rats in electroacupuncture group,traditional Chinese medicine group and acupuncture-medicine group was significantly lower,while the intestinal propulsion rate was significantly higher.The gastric residual rate in the acupuncture group decreased most significantly(P < 0.05),the degree of decrease in the traditional Chinese medicine group was also significantly higher than that in the electroacupuncture group(P < 0.05),and the intestinal propulsive rate in the acupuncture group increased the most,while the intestinal propulsive rate in the traditional Chinese medicine group was significantly lower than that in the electroacupuncture group(P< 0.05).5.After modeling,the contents of LP and CCK in serum of rats in FD model group,traditional Chinese medicine group,electroacupuncture group and acupuncture-medicine group were significantly higher than those in normal group.After intervention,the contents of LP and CCK in serum of rats in traditional Chinese medicine group,electroacupuncture group and acupuncture-medicine group were significantly lower than those in FD model group,and the decrease of serum LP and CCK in acupuncture-medicine group was the most obvious,but there was no significant difference between traditional Chinese medicine group and electroacupuncture group(P > 0.05).6.After modeling,the positive expressions of LP and CCK in stomach and brain tissues of rats in traditional Chinese medicine group,electroacupuncture group,acupuncture-medicine group and FD model group were significantly higher than those in normal group(P < 0.05).After intervention,the positive expressions of LP and CCK in stomach and brain of rats in traditional Chinese medicine group,electroacupuncture group and acupuncture-medicine group were significantly lower than those in FD model group,and the down-regulation of LP and CCK positive expression in stomach and brain tissue in acupuncture group was the most significant.In gastric tissue,the decrease of LP in traditional Chinese medicine group was greater than that in electroacupuncture group,while the decrease of CCK in electroacupuncture group was less than that in electroacupuncture group.In brain tissue,there was no significant difference in the decrease of LP and CCK between the traditional Chinese medicine group and the electroacupuncture group.Conclusion:1.The traditional Chinese medicine compound Weiyong granule combined with electroacupuncture at "Zusanli" and "Neiguan" points can significantly enhance gastrointestinal motility.2.Traditional Chinese medicine compound Weiyong granule combined with electroacupuncture at "Zusanli" and "Neiguan" acupoints can significantly down-regulate the contents of LP and CCK in peripheral serum and protein expression in stomach and brain tissue of FD rats.3.The therapeutic effect of traditional Chinese medicine compound Weixing granule combined with electroacupuncture at "Zusanli" and "Neiguan" on FD rats,suggesting that the combination of acupuncture and medicine can play a therapeutic effect on FD rats by regulating the secretion of LP and CCK and restoring the balance of BGA. |
