Uncaria rhynchophylla is an evergreen vine of the genus Uncaria in the Rubiaceae family.Dry hook stem can be used as medicine,the active ingredients are rhynchophylline,isorhynchophylline and hirsutine etc.,can act on cardiovascular and central nervous system diseases.However,due to the instability of active components,especially rhynchophylline and isorhynchophylline,they are easily lost during processing.The aim of this study is to reduce the loss of medicinal components by investigating the detection method and extraction process of rhynchophylline,isorhynchophylline and hirsutine in Uncaria rhynchophylla,as well as the drying method and the pressing process of Uncaria rhynchophylla to drink tablets.The main results are as follows:(1)Establishment of methods for the determination of rhynchophylline,isorhynchophylline and hirsutine in Uncaria rhynchophyllaHPLC was used to establish a method for the detection of rhynchophylline,isorhynchophylline and hirsutine in Uncaria rhynchophylla:Elite Hypersil ODS C18column(4.6 mm×200 mm,5μm);methanol-water(with 0.06%triethylamine,p H 7.3adjusted by glacial acetic acid)(74:26);injection volume 5.0μL;column temperature30.0°C;detection wavelength 245 nm.The linear relationship of the method was good,and the average recovery was 91.14%-114.08%.(2)The optimization of the extraction process of rhynchophylline,isorhynchophylline and hirsutine from Uncaria rhynchophylla by response surface methodOn the basis of single factor experiment,Box-Behnken model was used to conduct response surface method design to investigate the influence of ultrasonic time,ethanol concentration and solid-liquid ratio on the extraction effect of rhynchophylline,isorhynchophylline and hirsutine.The results showed that the optimal extraction conditions under ultrasonic time 53 min,concentration of ethanol 65%and solid-liquid ratio of 1:22 were recommended.(3)Drying characteristics and modelling studies on Uncaria rhynchophyllaIn this study,six common drying models were used to fit the drying curves of Uncaria rhynchophylla pieces at 40,50,60,70 and 80℃.Correlation coefficient(R~2),chi-square(X~2)and root mean square error(RMSE)were used as indicators to evaluate the degree of fitting,and the best drying model of uncaria decoction pieces was selected.The results of the study show that the drying model Page is the best drying model for Uncaria rhynchophylla.(4)Effect of different drying conditions on the quality of Uncaria rhynchophyllaBased on high performance liquid chromatography(HPLC),the contents of rhynchophylline,isorhynchophylline and hirsutine of Uncaria rhynchophylla at different drying temperatures and times were determined.To study the changes of the three active components in the drying process,and to screen the best drying method.The changes in the content of rhynchophylline,isorhynchophylline and hirsutine were found to rise and then fall under drying at different times at seven temperatures of Uncaria rhynchophylla,and the optimum drying temperature and drying time for Uncaria rhynchophylla was obtained as 1580 minutes at 55°C.(5)Optimization of the process for pressing the tablets of Uncaria rhynchophyllaBased on the theory of the existing tablet preparation process and technology,the influence of filling depth and filling pressure on the comprehensive score was studied by taking the color,shape,brittleness and tablet weight difference as the comprehensive evaluation index.Finally,the technological parameters of Uncaria rhynchophylla pressing decoction pieces were determined:the filling depth was 16 mm,the filling pressure was 90 k N and the rotating speed was 6.0 rpm.In conclusion,the HPLC method was established to determine the content of active components in Uncaria rhynchophylla effectively.The optimized extraction method is conducive to the utilization of alkaloids in Uncaria rhynchophylla.By optimizing the drying method and tablet pressing technology in the processing of compressed decanting pieces of Uncaria rhynchophylla,the contents of rhynchophylline,isorhynchophylline and hirsutine were retained to a large extent,and the utilization rate of Uncaria rhynchophylla was enhanced,which provided technical support for the extension of Uncaria rhynchophylla industry chain and had practical significance for the rational utilization of Uncaria rhynchophylla. |
