PDX-1 gene was cloned and constructed to the recombinant retroviral vector expressing PDX-1 and LEPCs (liver epithelial progenitor cells) cell lines that stably express PDX-1 To study the potential and mechanism of LEPCs transdifferentiate into pancreatic cells, through adding cell factors to the medium , changing induce culture conditions during in vitro culture. The effect of ameliorating streptozotocin-induced hyperglycemia after implanted LEPCs PDX-1 into diabetes mice was. The results indicated that LEPCs can transdifferentiate into islet-like cells and can express insulin granulas, it also can changed the symptoms of diabetes mouse. These results not only can be benefit to study the transdifferentiation mechanism of hepatic cells to pancreatic cells, but also supply important dates and theoretical according in cell therapy in diabetes mellitus.
