ANF(Atrial Natriuretic Factor) is a polypeptide hormone with potent natriuretic, diuretic and vasorelaxant effects which is also involved in the homeostatic regulation of fluid and electrolyte balance. These properties permit efficacious applications of ANF in a variety of clinical therapeutic uses, including essential hypertension, congestive heart failure, renal failure, edematous conditions and asthma, et al. Most of ANF used in clinical research were chemically synthesized by solid-phase method, which was costly. So it is an ideal way to produce cheap ANF in amounts by genetical engineering.α-hANF, a peptide composed of 28 amino acids with a disulfide linkage between 7 and 23 cysteine, is very unstable and susceptible to protease in E.coli cells when expressed in native form. The problem may be overcome by fusing the peptide to a leader sequence which is specifically removed later. The a-hANF gene, with the codon for Glu in its 5' end, was cloned into the expression vector pLSD13 at EcoRI-BamHI sites, resulted in the Ref--Glu-( α -hANF) fusion relationship.
