Introduction2,3,7,8 - tetrachlorodibenzo - p - dioxin (TCDD) is an environmental contaminant with the highly toxic effects, including teratogenicity, carcinogenic-ity, and mutagenecity. The mechanism of the reproductive toxicity exposed to TCDD is still unclear. In this study, we built a model of abnormal growth fetal rats with TCDD. We explored the relation of TCDD - induced abnormal development in rats to insulin - like growth factor 2 ( IGF2 ) gene expression as well as the methylation status. To gain further insight into the mechanism of the TCDD - stimulated abnormal expression of IGF2 gene, we detected the IGF2 expression of rat hepatoma cell line BRL - 3 A with treatment of TCDD. We also studied the molecular pathway that TCDD induced the expression regulation of IGF2 gene.Materials and methodsAnimals ,cells and materials health adult Wistar rats, rat hepatoma cell line BRL - 3 A, the reagents for RNA extraction and reverse transcription, rabbit anti rats IGF2 polyclone antibody, rabbit anti rats C/EBPα polyclone antibody, rabbit anti rats C/EBPβ polyclone antibody, reagents for real - time quantitative RT - PCR and Western blot, reagents for DNA extraction and bisulfite - modified DNA sequencing, reagents for Electrophoretic mobility shift assay (EMSA) and Chromatin Immuno - Precipitation Assay (ChIP), reagents for MTT assay and flow cytometry.
