Optics in life and health science is a new point of growth in modern optics. With the development of optical technology in modern medicine, it is known as one of the most effective solution to diagnose and cure diseases such as cancer, tumors etc. However, the principle of light transporting in biological tissues should be clearly understood before those optical technologies can be applied correctly. Unfortunately, this problem is not well solved up to now, especially for the quick determination of light distribution in biological tissues and proper description of the optical properties of biological tissues. As a result, we will mainly discuss those two problems comprehensively in this dissertation.To solve the problem of quick determination of the light distribution in biological tissues, we introduce the linear system theory into Tissues Optics to establish the system theory for light transporting in biological tissues. And the new theory is verified by numerical calculation and experimental measurement. Those contents compose the first part of this dissertation. The productions in this part can be summarized as below:Firstly, the linear-shift-invariant properties of light transporting in biological tissues are discussed based on the radiative transfer theory. And a system theory of light transporting in layered biological tissues is established, which is compared by Monte Carlo method. The results show that it can improve the computational efficiency more than 10~2 orders. And then, the system theory is applied to calculate the converging light transporting in biological tissues and the diffuse reflectance of biological tissues irradiated by collimated light source. The results show that it is an effective tool for quick determination of light distribution in layered biological tissues. And then, an experiment is designed to verify the system theory, the results show that the theoretical calculations are well matched with the experimental measurements.Secondly, to make the application of system theory for light transporting in layered biological tissues independents on accurately knowing the optical parameters. We develop a new method for determining the point spread function of biological tissues from experimental measurements. We call it deconvolution method. Whole arithmetic of the method is presented, and which is used to obtain the transmission point spread function of musculatures. The result is well matched with the Monte Carlo simulation.Thirdly, the angular distribution of light in biological tissues is very important to develop and optimize the optical diagnosis and treat technologies. As a result, the angular distributions of light in biological tissues are investigated comprehensively, while laser light is transporting in biological tissues.
