This thesis consists of two parts: first, we study the function of phosphorylated ERK1/2 and CREB in nervous system. Second, we clone the cDNA encoding the C2A domain of aczonin, express it in E. coli and prepare its antiserum.1, ERK1/2 play critical roles in cellular transformation (proliferation and differentiation), whereas the fact that ERK1/2 are abundantly expressed in nervous system suggests that ERK1/2 may play important roles in postmitotic neurons. To investigate distribution of the protein in central nervous system is the first step to elucidate its function in central nervous system, so we investigated distribution of phosphorylated ERK1/2 in mice brain. Compared with the wide distribution of ERK2 in rat nervous system, the distribution of phosphorylated ERK1/2 in mice brain is more limited. The phosphorylated ERK1/2 was present in the nucleus, cytoplasm, neurites. The presence of phosphorylated ERK1/2 in nucleus, cytoplasm, neurites suggests a role for ERK1/2 in phosphorylation and regulation of both cytoplasmic and nucleus targets.
