Three kinds of primary substance in clinical medicine, i.e., endogenous small molecules, nucleic acids, and proteins, were investigated in the thesis. The possibility of routine application of capillary eiectrophoresis (CE) covering in wider range of the clinical medicine analytical field was discussed in detail from the viewpoint of method development and application. A CE technique for the measurement of human serum sialic acids, a kind of tumor marker, was primarily established. NANA in the serum of 72 patients v..th malignant tumors and 30 normal adults were determined respectively by CE without derivation. The new developed method was sensitive and simple. This originality suggests that it may be developed into an effective method for clinical indication of treating tumor, such as early diagnosing, selecting and monitoring treatment, and making prognosis. A modified non-gel sieving buffer system has been developed for the first time It was used to separate the STR polymorphic markers GATA62A1 I, D4S 1563, and DSPP. Gene linkage analysis was performed for a Dentinogenesis Imperfecta type II (DGI- II) family in this system. Preliminary location of the nosogenic gene was carried out successfully. It is the foundation of clone of the nosogenic genes and might be widely used for gene diagnosis in the fijture. A novel method for the measurement of insulin-link growth factor I (IGF-I) with capillary zone electrophoresis(CZE) was developed. The molecular interaction between IGF-I and its polyclonal antibody was primarily studied with immunocapillary electrophoresis (ICE) and the binding constant was determined. The work shows the possibility of applying CE to immuno-analysis, which could be a new clinical analytical method for diagnosis, treatment and monitoring for the endocritic and metabolic diseases. All three parts of the thesis aim to meet the requirement of practical clinical medicine and to elevate CE for clinical diagnostics from concept to reality. It also lays the foundation for the wide application of Chip-CE to the clinical medicine field in the future.
