Objective: For the improvement of the bioavailability of Chinese Materia Medica(CMM) and forthe rational use of nanotechnology to CMM is a quite singnificant. Using Margarita, Realgar, andRhizoma Coptidis as the representability for the animal, mineral and plant Materia Medicarespectively, the methods of preparation, the powder token, physical and chemical characteristics,the total dissolution quantity, the pharmacokinetics of effective components in vitro, the mainpharmacodynamics, the toxicity of the nanometer dimension powder were studied in the presentthesis. The results supply the scientific basis on the preparation and utility of CMM nanometerpowder.Methods: Ⅰ. The preparation of Margarita, Realgar, and Rhizoma Coptidis nanometer powder:Nanometer powder was prepared by the circumrotatory colliding supersound airflow disintegrator.Some influencing factors such as the feeding speed, the water content in CMM, the particle size andthe liquid nitrogen were studied. Ⅱ. The research of nanometer powder's token, physical andchemical characteristic: The appearance, microstructure, the powder particle size and the specificsurface area, aerated density of Margarita, Realgar, and Rhizoma Coptidis nanometer powder werestudied. The change of the moisture absorption of Margarita, Realgar, and Rhizoma Coptidisnanometer powder was studied by determining the speed of moisture absorption, the percentage ofmoisture absorption, the relative critical humidity and the flowability. The main chemicalconstituents of Margarita and Rhizoma Coptidis in normal powder, ultra-micro powder andnanometer dimension powder were studied be means of HPLC or TLC.In addition the content ofAs2S2 and limit quantity of As2O3 in Realgar's were studied. Ⅲ. The research of disperse stabilitymethod: The effect on the nanometer powder's stability of Margarita, Realgar, and RhizomaCoptidis of eleven dispersants such as PGE, PVP, Tween-80, SDBS, CMC-Na, PAA, PMAA-Na etc.were studied. The best dosage of the dispersant was determined. Ⅳ. The dissolution research ofeffective constituents: Using amino acid, arsenic and brberine as the effective constituents of theMargarita, Realgar, and Rhizoma Coptidis respectively, the total dissolution quantity and thedissolution rate from normal powder, ultra-micro powder and nanometer dimension powder in vitrowere studied. Their dissolution kinetics was analyzed. Ⅴ. The research on pharmacokinetics: Thepharmacokinetics process was studied by ig of 60mg·kg-1 (Realgar) and 0.8g·kg-1 (Rhizoma Coptidis)in SD rat respectively. The blood samples were obtained by cutting the rats' tails at different time.Graphite furnace atomic absorption spectrometry was applied to determine the content of serumarsenic in Realgar and the HPLC method was applied to determine the concentration of serumberberine in Rhizoma Coptidis. The serum drug's concentration- time curve of berberine was dealtwith DAS ver1.0 software. Then pharmacokinetic characteristics were compared among the normalpowder, ultra-micro powder and nanometer dimension powder of Realgar and Rhizoma Coptidis toVI 珍珠,雄黄和黄连纳米粉体的制备与特性研究find some characters and the rules. The research on the main pharmacodynamics and thetoxicity: Bacteriostatic and anti-inflammatory experiment were conducted on the normal powder,ultra-micro powder and nanometer dimension powder of Rhizoma Coptidis. Realgar'pharmacodynamics experiment adopted antitumour method comparing the normal powder,ultra-micro powder and nanometer dimension powder. Antitumour experiments in vitro were used tomeasure the effect of blood serum which containing the nanometer dimension powder of Realgar toL1210 and K652.For the study on Margarita, the abirritation and sedation of the normal powder,ultra-micro powder and nanometer dimension powder were compared.
