With the fast development of biology and medicine, there is an urgent need for highly-efficient and sensitive analytical methods used for disease diagnosis and therapeutic drug monitoring. Amino acid neurotransmitter and protein cellular factor are not only important bioactive substances in organism, but also the characteristic substances associated with some neuropathology and cancer. Owing to the very low concentrations of amino acid neurotransmitters and protein cellular factor existed in biologic body and the complexity of sample constitute, the analytical methods for these substances must possess high-efficiency, high-sensitivity and good particularity. When chiral drugs are taken orally or injected into biological body, they are transferred to target organ, the determination of enantiomer concentration in biological fluid and the measurement of binding constant of drug with human serum albumin (HSA) have a great significance in the pharmacology researches and stereoselective pharmacokinetic analysis of chiral drugs.In this paper, capillary electrophoresis was combined with laser-induced fluorescence detection, immunoassay and field-amplied sample injection (FASI) technique. A highly-efficient and sensitive analysis method of the amino acid neurotransmitters in the cerebral cortex of rats was established. A competitive immunoassay based on capillary electrophoresis with laser-induced fluorescence has been developed for the determination of daintain/AIFl in human serum. The highly-efficient and sensitive enantioseparation methods for four basic pharmaceutical enantiomers i.e, fluoxetine, metoprolol, trihexyphenidyl and carteolol in biologic fluid were set up. The major contents in this thesis are described as follows:(1) A highly-efficient and sensitive analytical method of the amino acid neurotransmitters in the cerebral cortex of rats was developed by capillary electrophoresis with laser-induced fluorescence detection and fluorescein isothiocyanate (FITC) derivatization. Different parameters that affected derivatization and separation were studied and optimized. The separation of amino acids was performed using an uncoated fused-silica capillary (57 cm 75 m I.D.) with a buffer of 15 mM borate at pH 9.2 and an applied voltage of 17.5 kV. The detection limits for six amino acids were in the range of 2.1xl0-11M.-6.3x10-10M. This method was used to investigate the pharmacological effect of baicalin during cerebral ischemia. The results show that cerebral ischemia can cause a significant elevation of the concentrations of Glu, Asp, GABA, and Gly in cerebral cortex.Baicalin administration can attenuate the elevation of Glu and Asp induced by cerebral ischemia. This research demonstrates that baicalin may act as a neuroprotectant during cerebral ischemia.(2) A competitive immunoassay based on capillary electrophoresis with laser-induced fluorescence detection has been developed for the determination of allograft inflammatory factor 1 (daintain/AIFl), which is a bioactive macrophage factor. In this study, daintain was labeled with fluorescein isothiocyanate (FITC), and then FITC-daintain was purified using high-performance liquid chromatography. The experimental conditions affecting immumocomplex formation and CE separation were studied and optimized. The optimal CEIA operation parameters were as follows: 8.0X10~8 M anti-daintain, l.QxlO"7 M FITC-daintain and sample were mixed and incubated in PBS buffer at 25癈 for 25 min, then CE was performed using an uncoated fused-silica capillary (37 cm x 50 um I.D.) with a buffer of 67mM PBS buffer containing 0.05% hydroxypropylmethylcellulose at pH 7.4 and an applied voltage of 2 5 k V. Free FITC-daintain and daintain/anti-daintain complex could be separated within 7 min with high reproducibility. The detection limit of daintain was 3.4nM, mass LOD was 17amol.
