Constructing multi-components library (MCL) by integration of moderntechniques is an important way to accumulate chemistry and informationresources for continuous utilization in the research of Traditional ChineseMedicine (TCM). In order to develop efficient methods and techniques, whichare useful to construct library, the dissertation purpose for the first time thestrategy of separation of components from TCM along with the principle ofintegration of various tools. A separation scheme was designed, in whichcomponents could be mainly prepared by high performance liquidchromatography under the direction of analytical results acquired by usinghyphenated techniques. In the dissertation, methods were developed forseparation and characterization of different types of components, rapidqualitative analysis of micro components and construction of MCL.Guided by the separation strategy, lots of acid, basic and neutralcomponents were prepared from Strychnos nux-vomica. Thirty-two purecompounds, including fifteen new ones, were identified by spectroscopicmethods. By using on-line coupling of reversed phase highperformance liquidchromatography to tandem mass spectrometry, six minor hop acids, including''adprelupulone''found for the first time in natural resources, were detectedtogether with the six major ones even lack of reference compounds. Methodsof rapid preparation, chemical characterization and sample managementwere studied in the construction of hormone MCL. The difficulties and keypoints in the course of construction were discussed.Successful preparation of micro components showed the practicability ofthe established separation model. Meanwhile, the research highly enricheschemical information of the studied Chinese medicines. The methods of linearscale-up and dynamic incision expand the application of preparative highperformance liquid chromatography for rapid separation of differentcomponents. The research demonstrates that it is feasible to construct MCLby integration of the related methods and techniques.
