Studies On The Effects Of Two Herbicides On Anabaena Flos-aquae & Superweak Luminescence

With the development of social economy, waterbody entrophication has become a serious environmental problem and have aroused the attention of many people. The harm caused by water bloom and red tide was frequently reported, which has prevented the development of society. However, countermeasures to controll the freshwater and red tide have not been taken yet. Anabaena flos-aquae is one of blue algae which leads to the birth of water bloom. The effect of Metsulfuron-methyl and Bensulfuron-methyl on the growth of algae was discussed in this paper. Visible Spectrophotography was employed to measure the Index of Absorbance which can be viewed as an index of the standing crop of Anabaena flos-aquae. We have tried the experiments in small-scale body of water and measured the amount of Metsulfuron-methyl and Bensulfuron-methyl residue in algae suspensions. At the same time, the mechanism of Metsulfuron-methyl on Anabaena flos-aquae has been preliminarily studied by observing the sensitivity of acetolactate synathase(ALS) to Metsulfuron-methyl in vitro and in vivo respectively. The ultraweak bioluminescence and delayed bioluminescent spectrum of Anabaena flos-aquae have been first systematically detected. The main results show as follows:1 Culture of Anabaena flos-aquae in laboratoryAnabaena flos-aquae were inoculated in 200mL SE culture medium under germ-free condition and cultured at 25 ± 1 °C with illuminzation of fluorescent light (4500lux, 24h/day). Air was blown into algae suspension with air-pump. The cultivation continued under the same conditions as mentioned above. This assured suffcient algae and made the trial continuous.2 Purification and token of Metsulfuron-methylThe powder of 10% Metsulfuron-methyl was purified with acetone: dichloromethane (1: 1)by extraction, centrifugal, filtration, dryness and recrystallization. The sample purified by myself and the standardized substance bought have the same melting point. They have similar purity also. Their gas chromatography and NMR are almost identical. So the refined Metsulfuron-methyl can function as a standardized substance which is cheap and convenient.
