Chrysopa septempunctata Wesmael is a kind of important natural enemy insects, which can control agricultural and forestry pests effectively, such as aphids, coccids and aleyrodid. Due to wide feeding habits, large appetite, wide distribution and large quantity, it has been paid great attention to by biological control workers. At present, research has been carried on about resources of green lacewing and its biology, ecology, ethology of common species, and reproductive technology on a large scale, natural feed and artificial feed as well. However, research on reproductive physiology respect of green lacewing now is less and hasn't gone too far. This article has carried on certain studies on this respect, aiming at offering theoretical direction for giving play to green lacewing's reproductive latent, raising artificial reproductive efficiency and field effective protection and utilization. The main conclusions are as follows:The vitellin of Chrysopa septempunctata was extracted with 0.4mol/L NaCl and then precipitated in cold distilled water. Ion exchange chromatography and proteic gel recovery were used to purify it further. SDS-PAGE analysis shows that the vitellin is composed of two subunits, the large one 165kDa and the small one 46kDa.Then automatic amino acid analyzer was used to analyse amino acid composition of vitellin.A polyclonal antibody against vitellin was prepared in male rabbits and the specific antibody against vitellin was made through absorbtion by male worm homogenate. After purification, HRP was used to mark antibody, then ELISA was set up to detect vitellin content in the Chrysopa septempunctata fat-body, hemolymph and ovary.Chrysopa septempunctata was fed with two foodstuffs, aphids and artificial ovum Trichogramma's pupas. The results indicated that survival time of female worms which intook aphids was obviously longer compared with those which intook artificial ovum Trichogramma's pupas, and their ovum quantity obviously rose. But artificial ovum Trichogramma's pupas can be used as supplementary foodstuff to raise green lacewing because of its factory production. Through ELISA detection, vitellin's synthesis and intake of female worm taking in artificial ovum Trichogramma's pupas were both obviously reduced.Temperature is an important environmental factor which influences insects' reproduction. In order to obtain the highest reproductive efficiency, Chrysopa septempunctata was fed with aphids at 15 # ,25 # and 30 # respectively.The results indicated that oviposition period was prolonged obviously and oviposition quantity to a single female worm was the highest at low temperature of15#. Through ELISA analysis, the time of vitellogensis was prolonged relatively in fat body, haemolymph and ovary.The titer variation to juvenile hormone of female worms was detected by GC-MS system, from which there was an obvious correlation between variation regularity to juvenile hormone and vitellogensis's regularity in Chrysopa septempunctata.The variation to vitellin polypeptide was studied by SDS-PAGE during embryogenic and postembryonic development to Chrysopa septempunctata. The results indicated that vitellin was degradated constantly during embryonic development, and the bands of vitellin couldn't be detected in the 4th day of ovum development. The content variation to Chrysopa septempunctata's vitellin was detected through ELISA. It indicated that vitellin was utilized gradually during embryonic development, and a fallowing trend in form of straightline was found in the whole embryonic development. Vitellin couldn't be detected in just hatched larvae. The most obvious variation to ovum morphological character was that the color was changed from light green to sepiaceous in embryonic development, and all the ovums were almost hatched after 96h.According to amino acid sequence to vitellin which has the structural character of GL/ICG motif near C end, two degenerate primers were designed and Chrysopa septempunctata 's RNA was extracted, and a 900bp fragment was amplified by RT-PCR.
