Drought is one of important abiotic limiting factor for yielding wheat (Triticum aestivum L.). It is of great importance to study the mechanism how plants respond to and against water stress, which is helpful to find out the genetic foundation of drought resistance, discover new genes concerned about drought resistance and breed crops against drought. To investigate drought resistance mechanism in wheat, we used several techniques including cDNA library construction of suppression subtractive hybridization (SSH), differential hybridization screening (cDNA Macroarray) by high density dots membrane and bioinformatics methods.Hanxuan 10 (Jinmai 5) was used, which is an elite wheat cultivar from 20000 with strong drought tolerance. The seedlings were cultivated in polyethylene glycol 6000 (V=-0.5MPa) solution for 12h, 24h and 48h, respectively. A localization platform was constructed for EST sequence analysis. Based on Linux operation system, the platform consists of perl script and other free soft wares. It could be batch done with Blast, EST annotation and function classification. Thus this platform will be useful to know the bioinfonnation of EST sequences of wheat in drought resistance and the expression patterns of genes related with drought resistance.Three cDNA libraries were constructed using SSH, which included wheat seedlings under water stress for 1h, 6h and 12h, respectively. 1697, 1131 and 1833 ESTs were respectively obtained from these libraries and sequenced. Sequence assembly revealed that 938, 566 and 516 contigs were acquired. 114, 203 and 133 Uni-gene were ultimately found out using blast, function annotation and classifications. These data laid a solid foundation for analyzing gene profiling of wheat in water stress.The expression modes of the genes responding to water stress in wheat seedlings were obtained.
