Beijing ducks is a famous breed of ducks.It plays important roles in ducks breeding and ducks industry development in the world. But the developments of breast muscle of Beijing ducks have two characteristic. The breast muscle rate was low and the development of breast muscle was relative late.In order to improve the development of Beijing duck's breast muscle.we researched the expression of MSTIN, GHR, IGF-1, MyoG in brest muscle and the relative analysis of the polymorphism candidate gene with the candidate index of breast muscle .On the other hand, GHRH gene was cloned.The resules were as followes:In our study, 550bp partial cDNA of Myostatin (MSTIN) gene (Genbank accession number DQ168594 ),345bp partial cDNA of growth hormone receptor (GHR) gene (Genbank accession number DQ137140 ) and 507bp partial cDNA of Insultin-like growth factor-1 (IGF-1) gene (Genbank accession number DQ168593,) were cloned form RNA isolated from Beijing duck's breast muscle. The duck MSTIN cDNA coded for a predicted protein of 182 amino acids and deducted amino acids of partial GHR cDNA, IGF-I partial cDNA were 124 and 182,respectively.Genbank accession number of deducted amino acids were ABA02292, ABZ50386 and ABA02291,respectively. Myogenin (MyoG) extron 1 partial sequence was cloned, also. Nucleotide sequences and deducted amino acid sequences were enrolled in Genbank. Genbank accession numbers were DQ137140 and AAZ72963. The results provided a major base for researching gene expression, cloning complete CDS and polymorphisms analysis of duck MyoG, MSTIN IGF-1 and GHR. On the other hands, it was necessary to study the development of change of these gene expressions in the Beijing duck's breast muscle and search for the effect of IGF-1 to the breast muscle development of Beijing duck.In order to search for the effect of IGF-1 to the breast muscle development of Beijing duck, we studied the development of change of candidate gene (including MSTIN, GHR and IGF-1) expression in the Beijing duck's breast muscle. The results showed that MSTIN expression in breast muscle was higher at 28, 35 and 42 d than at 7, 14 and 21 d. The pattern of MSTIN expression was closely parallel to the trend of breast muscle growth, suggesting that MSTIN might play an important role in breast muscle development. It was possible to postulate that MSTIN may be a major determinant of muscle mass in breast muscle, as shown in other species.IGF-1 mRNA expression abundance in the Beijing duck's breast muscle was not significantly different at the age of 7d,14d,21d and 28d,35d,42d, respectively. But, IGF-1 mRNA expression abundance in the Beijing duck's breast muscle at 28d, 35d, and 42d of age was higher than that of at 7d, 14d, and 21d of age.
