F1 and F2 offsprings population were established by grape interspecific hybridization from 2001 to 2005. The resistance to Uncinula necator and Sphaceloma ampelinum of Chinese wild Vitis, European grapes (Vitis vinifera L.), American native grapes and the hybrids was identified respectively, and then genetic demonstration of the resistance to Uncinula necator and Sphaceloma ampelinum of Chinese wild Vitis was analysed. Additionally, gene markers and their application of assisting breeding selection and sequence analysis of disease-resistance genes were performed in order to map and clone the genes for future research. The results are as follows:1. The results of identification of the resistance to Uncinula necator of grape germplasm resources indicate: 11 clones of 9 species of American native grapes are all resistant to Uncinula necator, among which the resistance of Alachua and Conqurster of V. rotundifolia, A. De Serres of V .rupestris and V. berlandieri is highest; 14 cultivars of V. vinifera L. are all susceptible to Uncinula necator, among which the susceptibility of May Purple is feeblest. Baihe-35-1 of 9 clones of V. pseudoreticulata is the most resistant to Uncinula necator, then Baihe-13, Baihe-13-1 and Guangxi-1. Baihe-35-2, Guangxi-2, Shangnan-1, Shangnan-2 and Hunan-1 are the susceptibility. Hunan-1 is the most susceptible to Uncinula necator.2. The results of identification of the resistance to Uncinula necator under natural condition indicate: The resistant inheritance of Chinese wild Vitis in which existed major resistant genes were controlled by polygenes, showing the heredity of quantitative traits. The resistance of major and minor resistant genes has the accumulative action in hybrid offsprings. The heredity of RAPD and SCAR markers of disease-resistance genes between interspecific hybrids F1 and F2 generation also indicate: There existed many major resistant genes in Chinese wild Vitis.3. 5 RAPD markers and 1 SCAR marker (OPO11-964,OPY13-661,OPW02-1756,OPB11-520,OPW05-766 and SCO11-914; Nucleotide sequence data are available in the GenBank databases under the accession numbers DQ493890,DQ493892,AY491396,AY491398 and AY491397) of the resistance to Uncinula necator of Chinese wild Vitis, and 1...
