Fruit shape is one of the most important characters in cucumber. In production process, the shapes of most fruits are anomalous. A majority of anomalous fruits are band fruits. Band fruits make character, smell and taste of fruit bad. Band fruits also make the effect of cucumber as sale crop. So it is significant to reduce the rate of band fruits for cucumber breeding and cultivation. It is necessary to definitude genetic affinity, do genetic analysis, search pertinent special proteins and investigate genetic mechanism on cell and molecule level for researching the character at the point of genetic breeding and making bend prevented or cured. Studies on bent fruit and molecular- assisted breeding conduce to make the number of bent fruit decreased, enhance fruits value and significantly provide theory gist for nail down genetic mechanism and cultivate new variety with excellent fruits in cucumber.We found a new precision, science and high frequent method for measuring camber of fruits in cucumber during MATLAB language. This method make veracity and efficiency heightened markedly. We make bent variety and straight variety as parents for genetic analysis in the experiment, mark F2 separate population and QTL mapping, research genetic mechanism. We abstract pertinent special proteins of representative part and doing mass spectrometry during 2D-PAGE and analysis internal mechanism. We research the bent trait during associated computer image recognition technology with genetic analysis on proteomics studies level. The main results were as followed:1. Exploit a series of modules about image collection, grey level, dividing up fruit and background (including grey level of pictures, format transformation and binaryzation), marginal check, contour extraction and so on during MATLAB language. Establish exact and frequent picture recognition system on the shape of cucumber fruits.2. To search the mid-axes of cucumber fruits using genetics arithmetic, accommodated function is as follows:The parameter are as follows: population=100, across probability=0.8, variation probability=0.02. 3. The result shows that bent character of cucumber fruit is quantity genetic controlled by multiple gene and controlled by major gene. The additive effect plays the main role, and dominance effect plays the lesser role. Compared with the additive-environmental variance, the dominance- environmental variance is more important and cucumber fruit is more easily affected by additive effect. Broad heritability and narrow heritability are both higher in bent character of cucumber fruit and is separate 48.447% and 53.122%. The variety of D0455,D07299 and D0422 can be used as parent which are benefit of improving the straight character of cucumber fruit. And that the bent character of cucumber fruit in L18-10-2 is transmitted to filial generation maximumly.4. 7 pairs of SSR primers were screened out during the parent and the DNA mixing pool of the progeny.CSTCC813,CSWCT27,CSWCT29,CSWACC02,CSWTA08A,CSWATT02 and CSWCT25. Thereinto CSWCT27,CSWCT29,CSWACC02 and CSWATT02 belong to the same molecular marker linkage group, the distance of two markers separate is 12.1cM,17.5cM and 18.2cM. The length of linkage group is about 47.8cM.5. By the software of Windows QTL Cartographer V2.0 and the method of composite interval mapping, one QTL was detected for bent fruit of cucumber. The distance between QTL loci and marker were 2.5cM. It can explain genetic variability 9.33%.6. Optimize the method of distilling the protein of cucumber fruit for 2D-PAGE. The key is taken by following aspects. To skive template using MM301 cell break instrument at 30Hz for 3min, cannot put in PVP in this progress. The optimization value is seven times to pure protein during acetone again and again. The optimization value is 40mg/mL as pattern dosage of IEF.7. During 2D-PAGE, 32 difference proteins from different part of cucumber fruit are screened out. Ten of them are taken on mass spectrometry. The result from MASCOT software shows that three of ten proteins are some a kind of big subunit of ribulose-1, 5-bisphosphate carboxylase/oxygenase large subunit, others are isocitrate dehydrogenase,Cobalamin-independent methionine synthase isozyme,14-3-3 protein and GTP-binding protein, a unknown protein.
