Objective Catechol-O-methyl transferase (COMT) is a promising candidate gene for schizophrenia. A non-synonymous single nucleotide polymorphism (SNP) (rs4680) in COMT has been investigated for its possible association with schizophrenia in the Han Chinese population many times, however, the results are inconsistent. Moreover, the effects of COMT on brain structure variations in the Han Chinese population remain still unclear.Methods Here, we performed a meta-analysis of rs4680 using all available Han Chinese case-control subjects, including a total of 4,740 cases and 5,323 controls. We also tested the associations of three SNPs (rs4680、rs165599、rs737865) with brain structure volumes in a healthy Han Chinese imaging sample (N=299).Results Our meta-analysis results do not support an association of rs4680 with schizophrenia (P=0.51).Rs4680 shows nominally significant association with the volume of caudate nucleus, especially in male subjects. In female subjects, rs4680 is significantly associated with the volume of middle occipital gyrus and insula, rs 165599 is significantly associated with the volume of insula and thalamus, rs737865 is significantly associated with middle frontal gyrus and inferior temporal gyrus. In male subjects, rs737865 shows nominally significant association with the volume of postcentral gyrus.however, these results could not survive multiple corrections.Conclusion Although our results suggest that rs4680 in COMT is not a risk SNP for schizophrenia in the Han Chinese population。Rs4680、rs165599、rs737865 may play pivotal roles in some brain regions, such as the caudate nucleus, or affect brain development, and these points should be investigated in the future.Objective:rs2312147 is a novel single nucleotide polymorphism (SNP) in vaccinia virus kinase related kinase 2 (VRK2) who has been reported significant association with schizophrenia and significant effects on total brain volume in a Chinese sample. But the situations on the brain regions related to schizophrenia were still unknown.Methods:Here, we tested the associations of the SNP with 19 brain regions volumes involved in the etiology of schizophrenia in a healthy Chinese imaging sample (N=299).Results:The results showed that rs2312147 was significantly associated with the volumes of multiple brain regions, with the risk allele indicating smaller volumes. Especially the caudate nucleus and the middle occipital gyrus volumes remained significant after Bonferroni correction across multiple brain regions.Conclusion:So our brain imaging data suggested a potential role of VRK2 in brain structure and function, implying a possible pathophysiological mechanism underlying schizophrenia susceptibility.I Objective The single-nucleotide polymorphism (SNP) rs1344706 located at gene ZNF804A (zinc finger protein 804A) was identified as the first genetic variant achieving genome-wide significance for schizophrenia.Methods Here, we tested the associations of the risk SNP with 19 brain regions volumes involved in the etiology of schizophrenia in a healthy Chinese imaging sample (N=299) to analyze the biologic function of the gene ZNF804A and the neural mechanisms of risk.Results The results showed that rsl344706 was not associated with the volumes of any brain regions.Conclusion So our brain imaging data suggested ZNF804A rs1344706 has no effect on brain structure and more likely affect functional connectivity and leads to the schizophrenia susceptibility.
