Part ⅠTo investigate the relationship between serum 25(OH) VD levels and childhood asthmaObjective: 1. To investigate the relationships between the incidence of childhood asthma and vitamin D levels;2. To analyse the relationship between disease severity and vitamin D levels in children with asthma.Subjects and Methods:The subjects were 100 patients with asthma, and 100 cases of normal control group. Diagnosis of asthma and period and severity had been made.Lung function was measured(including FEV1 absolute value, FVC and FEV1 / FVC)and food frequency surveys were made and relevant information was collected(age and so on);Blood samples were collected. The serum 25(OH) VD concentration was measured by enzyme-linked immunosorbent assay, and analysis of cellular immune function was made by flow cytometry. Statistics: General information and des-criptive data preclude the use of the t test and chi-square test; comparison between groups preclude the use of non- para- metric Mann-Whitney U rank sum test. Analysis of the relationship between25-OH-D levels and the risk of asthma in children was made by Logistic regression, and correlation between serum comparative tube 25-OH-D levels in children with asthma and various immune indicators was analyzed by multiple linear regression.Result:1.The 25-OH-D levels in asthmatic was 20.84 ± 6.80 ng /ml and that was 28.83 ± 6.69 ng / ml in the control groupe,and.There was a significant difference.(t = 44.875, P <0.001).2. The serum 25-OH-D levels in asthma and control groups in the spring was statistically significant(χ2 = 8.618, P <0.05), and so was in summer(χ2 = 4.612, P <0.05), and so was in fall(χ2 = 13.134, P <0.05),and so was in winter(χ2 = 16.915, P P <0.05).3.The difference of 25-OH-D levels between mild persistent asthma group and moderate persistent group was not statistically significant(χ2 = 72.155, P >0.05), and those between moderate persistent and severe persistent group was not statistically significant(χ2 = 43.770, P >0.05). But those between mild persistent group and severe persistent group was statistically significant(χ2 = 62.000, P <0.05). 4. The serum of 25-(OH)-VD was positively correlated with lung function(p <0.05). 5. The serum 25-(OH)-VD concentration was positively correlated with CD4, CD4 / CD8.Conclusion:1. Serum vitamin D was lower in asthma group than the control group, and it may be associated with childhood asthma;2. Serum vitamin D levels was negatively correlated with asthma severity;Part ⅡTo study the association between gene single nucleotide polymorphisms and asthmaObjective 1.To explore associations between single-nucleotide polymorphisms of CYP27B1, GC, VDR gene and childhood asthma; 2. To analyse the relationship between disease severity and single-nucleotide polymorphisms of CYP27B1, GC, VDR gene in children with asthmaSubjects and Methods The subjects were 100 patients with asthma and 100 cases of normal controls.Blood specimens were collected from all subjects and stored frozen. Genomic DNA of subsequent were reunification. Gene single nucleotide polymorphism genotype and allele frequencies were detected by Sequenom technology in Mass ARRAY system and the relationship between the children and asthma was studied, and further analysis of asthma severity, lung function and single nucleotide polymorphism correlation were undertaked. Statistics: SNP per locus genotype and allele frequency was tested by chi-square to detect whether the difference in the two groups was significant; the use of multiple independent samples nonparametric test to detect asthma locus and disease severity and lung function if there is a correlation.Results1. The distribution of genotype GA in VDR gene locus rs1544410 between asthma group and control group was statistically significant(χ2 = 1.22, P <0.05).2. GT genotype of CYP27B1 gene locus rsl0877012 in the asthmatic group was significantly associated with severe asthma(P <0.05).Conclusion1. Genotype GA of VDR gene rsl544410 was associated with childhood asthma;2 Genotype GT of CYP27B1 gene rs2282679 was closed related severe asthma;...
