Objective mRNA differential display assay was developed to compare gene expression between M. Tuberculosis H3?Rv and its avirulent mutant H37Ra.Methods Total RNA was extracted from M. Tuberculosis H3?Rv and HavRa that had been cultured 10-14days were used in the diiferential display. This analysis yielded some positive differential bands. These positive differential bands will be reamplicated, and sequence analyze and query homologous. Results This analysis yielded 10 differential bands, sequencuing 2 bands and query homologous for the bands in the NCBI database of GenBank. The bands were found highly homologous to M. Tuberculosis Hi?Rv genes. Conclusion These results show that mRNA differential display can represent a potential tool for investigation of M. Tuberculosis gene expression and can find virulence genes of M. Tuberculosis.
