Objective: To examine the influence of TNF a on thecontraction and [Ca2"]i of colon circular smooth muscle cells(CCSMCs) , and study the signalpathways involved in TNFa's effect.Method: bymeasuring the length of smooth muscle cells and the change of intracellular calcium , the effect of TNFa on the length and [Ca2+] i of CCSMCs , the response of smooth muscle cells to KCL and ACH and the influence of PD-98059 and manoalide on TNFa'seffect,with or without TNF a ,were observed. Result: The present study demonstrated that TNF a can't directly contract CCSMCs and have no influence on [Ca2+]i,but TNF a can increase the calcium sensitivity of CCSMCs to ACH,and PD-98059and manoalide partly repress TNF a 's effect. Conclusion: This study demonstrates the ability ofTNF a to enhance the calcium sensitivity of CCSMCs to ACH, which was shown to be mediated by ERK MAPK signal pathway and PLA2.
