[Objective] ANFH induced by steroid or alcoholism are the most frequent etiological factor in the non-traumatic ANFH, which contained 2/3 of ANFH and its change of histology and pathophysiology are very similar, that may indicate congenerous pathogenesis. Today, the effect of endogenetic GC in the pathogenesis of ANFH by alcoholism is not understanded, although the endogenetic GC increased when overdoes drinking had been confirmed. To study the effect and to explore the pathogenesis of endogenetic GC in ANFH by alcoholism, we poured hight dose distilled spirit into stomach of rabbits, the others were administered isodose distilled spirit and inhibitor of GC. It would lay a foundation and provide the theory and experiment data for prophylaxis and therapeutics for ANFH by alcoholism.[Method] 90 New Zealand white rabbits were chosen and randomly divided into three groups. The experimental group were administered the strong distilled spirit containing 50%(v/v) of alcohol at dosage of 8mL?kg-1?d-1, the intervention group were given by isodose alcohol and aminoglutethimide 25 mg?kg-1?d-1, and isodose NS as controls by pouring into stomach with a plastic tube for 1 to 6 month,to measure serum cortisols by chemiluminescence. The animals in three groups were sacrificed in batches 1, 2, 3 and 6 months after the procedures. The serolog and hemorheology werestudied. SOD, MDA and ET in the homogenate of bone tissue were detected. The changes of histology in the hepar and femoral head were observed. The expression of GCRmRNA was studied by hybridization in situ. The osteocytes apoptosis was measured by means of TUNEL. The expression of p53 and Bcl-2 proteins by immunohistochemistry. [Results] General condition as follow: the animal of diet and action in the experiment and intervention group reduced, the response was apathy, the hair was dim, all animal showed malnutrition calorie and a few animals were digestive tract bleeding after pouring into stomach. BW growed slowly, the experimental or intervention group was significantly lower than control after third month (P<0.01), but no significant difference was found between the experimental and intervention group (P>0.05).The changes of serum cortisols in experimental group(5.48±1.33 ug?dL-1)>intervention group(2.14±0.56 ug?dL-1)>controls(0.73±0.24 ug?dL-1), wich found significant difference among the three groups(P<0.01).The expression of GCRmRNA in the osteocytes of the femoral head as follow: the experimental group expressed thinly in second month,thinly or negatively in third month and negatively in sixth month. the intervention group were secondary expression in first to third month and thin in sixth month, the controls were middling in first to sixth month. The change of serolog: the experiment and intervention group presenced hepatic functional lesion as GGT in second month, AST and ALT in sixth month, which increased remarkably compared with controls respectively. But no significant difference was found between the experimental and intervention group (P>0.05). TG inthird and sixth months, experimental group(2.65±0.81mmol?L-1,6.29±1.63mmol?L-1), intervention group(1.92±0.56 mmol?L-1,4.07±1.48mmol?L-1) and controls(1.16±0.34 mmol?L-1,1.29±0.52mmol?L-1), respectively, wich revealed brightly difference among the three groups (P<0.05,P<0.01). CHO showed significant difference among experimental group(2.79±0.45 mmol?L-1), intervention group(2.17±0.58 mmol?L-1)and controls(1.64±0.25 mmol?L-1)in sixth month(P<0.01).Hemorheology : In sixth month, respectively, BV at high, median and low shear showed significant difference among experimental group(5.96±0.65mpa?s, 12.60±1.26 mpa?s, 11.89±1.39 mpa?s), intervention group(4.91±0.84 mpa?s, 9.82±0.92 mpa?s, 10.12±1.93 mpa?s) and controls(4.06±0.67 mpa?s, 5.18±0.81 mpa?s, 8.68±1.80 mpa?s)(all P<0.01).
