part oneThe Culture and Inducing Differentiation of MBA-1 CellsObjective: To determine whether BMP-4 can induce MBA-1 cells differentiation into osteoblasts, and whether MBA-1 cells have multi- differentiation potentials.Methods: MBA-1 cells were induced by BMP-4 for 15-22 days, then HE staining, van Gieson's staining and alizarin red-s staining were performed to identify the osteoblastic phenotypes,with a positive control of mouse osteoblast precursor MC3T3-E1 cell lines.Results: As a positive control, MC3T3-E1 cells synthesized more type 1 collagen, and formed classical mineralized nodes regardless of the induction of BMP-4, but by the induction of BMP-4, MBA-1 cells synthesized more type 1 collagen, and more calcium deposition,without formation of classical mineralized nodes. A few cells enlarged in size and cytoplasm were filled with vacuoles.Conclusion: MBA-1 cells may have multi-differentiation potentials, BMP-4can induce the differentiation of MBA-1 cells into osteoblast-like cells.
