Fluorosis is one of the most seriously endemic diseases that was harmful to the health of people. It is popular in our country, there has been more that 0.1 billion people suffering from it so far. The mechanism of fluorosis is not clear now, and there is not effective drug to cure it. So it is very important to study its mechanism and to finding antagonizing factors.Fluorosis is a chronic cumulative poisoning that people lived in a high level of fluoride environment for a long time or intaked excessive fluoride, it mainly damaged rigidity tissue such as tooth and bone. In recent year, no - bone tissue injury of fluorosis has raised scientist attention in all over the world. The mechanism of fluorosis is complex, and even no accordant explain. Three theories were put forward by some researcher. 1) low nutrition and low calcium 2) free radical 3 ) magnesium theory.In recent year, people often consider that no - bone tissue injury by fluoride was connected with free radicals, because the theory of free radical was studied further. Many studying showed that fluoride can make rat or mice peroxidant much strongly, and increase MDA level, decrease the activity of antioxidant enzyme; supplementing with some extra antioxidants can antagonize or decrease peroxidantion resulted from fluoride. But there were some contradiction results. These results were due to differ in experiment condition or using methods, or reflect the diversity of mechanism of fluoride toxicity, it was difficult to conclusion.Whether fluoride was a latency carcinogenicity had been a topic of study the health of fluoride. The researcher had studied heredity toxicology of fluoride for20 years, and verified that fluoride can damage germ plasm, microkernel and chromosome aberration of cultured lactation cells including body cells. But there were some opposition results. The mechanism of fluoride damaging heredity material had not been clear.It is well known that many factors can induce body peroxidation such as hunger, tired, lack of oxygen, hurt, smoking and nutrition et al, and peroxidation can also result in injury of many tissues and organs of body. How about was the relationship between fluorosis and peroxidation? We should study their time order, universality, the intensity of their correlation in order to resolve the cause - result relation of peroxidative injury and fluorosis. It is benefit to determine biomarker of peroxidation and interdiction or protective experiment of tissue injury.Vc, VE, Se and GSH are important antioxidants of body, but the mechanism of their antioxidant are different. Therefore, we selected the rat cells of liver and kidney as target, studying the toxicity of fluoride that resulted in cells damaged through cell proliferation response firstly, and then determined its toxicity dose; studying the effect of fluoride on cells oxidative injury and DNA under this toxicity dose, and at same time to study the effect of different antioxidants on antagonizing cells damaged by fluoride, and screen out the best antioxidant antagonizing fluoride, we can provide scientific data in order to study the mechanism of fluorosis, prevent and treatment its happening and development.Material and methodsMaterial: NaF, Na2SeO3, a - Tocopherol ( E. Merck), GSH, Vc, RPMI 1640 medium (sigma, supplement with 10% fetus serum, 100U/mL penicillin, 100ug/mL streptomycin) , low melting point agar, normal melting point agar, TritonX - 100(sigma) , Tris, plain electrophoresis slot and electrophoresis instrument, Olympus BX60 fluorescence microscope (Japan).Methods: Using cultured liver and kidney of rat in vitro. Selecting growth well cells. Dividing them into two groups. NaF group: adding different levels offluoride into cells, cultured 12 hours. Antioxidant groups: Adding a certain level of Vc, VE, Se and GSH into cells, cultured 12 hours firstly, then cultured together with fluoride for 36hours.
