Studies On The Experimental And Clinical Effects Of Qi-Boosting And Suspended Rheum-Dispersing Beverage On The Local Immunologic Function And The Lesion Of Hydrothorax Complicated From Lung Cancer

This study was aimed at further confirming the therapeutic effect of Qi-Boosting and Suspended Rheum-Dispersing Beverage (QBSRDB) on hydrothorax complicated from lung cancer and at exploring its mechanisms for the treatment of this lesion . It was composed of following trials.1. Experiments on the Anti-tumor Effects of QBSRDB Here, cells of Lewis lung cancer and EAC cell line were transplanted to C57L/6J mice and Kunming line mice respectively to produce transplanted tumors among the animals, and the animals were fed with the solution of QBSRDB following transplantation of tumor cells. In this experiment, investigated in vivo were the effect of tumor-inhibiting on mice with transplanted tumors of Lewis lung cancer and the effect of survival duration-extending on mice with ascites due to transplanted EAC.2. Experiments on the Effects of QBSRDB on the Killing Activity of Tumor Infiltrating Lymphocytes (TIL) to Auto-Tumor Cells (ATC) In these experiments, TIL and ATC were separated from hydrothorax of cases with lung cancer and cultured for a period of time at first. Then, these cells were co-cultured in a media mixed with the solution of QBSRDB at an effector-target ratio of 40:1 for TIL to ATC. Moreover, these two kinds of cells were also separated from the ascites of animals with EAC transplanted and then, cultured in a media without QBSRDB solution, distributed at an effector-target ratio of 10:1 for TIL to ATL. Here, the killing activity of TIL to ATC was investigated through this experiment.3. Observation on the Effects of QBSRDB on the Levels of Cytokines Enzyme-linked immunosorbent assay (ELISA) was taken to determine the levels of IL-2/IL-2R, IFN-γ, IL-4 and IL-10 in the hydrothorax of patients with lung cancer and the levels of IL-2, IFN-γand TGF-β1 in the ascites of mice with EAC transplanted, all being tested before and after giving QBSRDB respectively.4. Qbservation on the Apoptosis-Inducing Effect of QBSRDB on Transplanted Lewis Lung Cancer In this experiment, two groups of amimals with Lewis lung cancer transplanted were studied to observe the apoptosis-inducing effect of QBSRDB on tumor cells, one group taken as controlling one without QBSRDB given and the other as treating one with this medicine orally taking. The tissue samples were investigated withimmunohistochemical assay to display the apoptotic cells and a comparison was then made between the two groups to see the apoptosis-inducing effect of this medicine on the transplanted tumors.5. Observation on the Effects of QBSRDB on the Expressive Levels of IL-2 and TGF-β1 Genes in the Cells of Lewis Lung Cancer As the same in the experimant stated as above, tissue samples were taken from the animals with transplanted Lewis lung cancer . These animals were divided into two groups, with one group being treated with QBSRDB and the other one taken as control. Then, RT-PCR method was used to determine the expressive levels of IL-2 and TGF-β1, and a comparative analysis was made to see the expressive difference of these two genes between them.6. Clinical Trial to investigate the Therapeutic Effect of QBSRDB on Patients with Hydrothorax Complicated from Lung Cancer In this trial, two groups of cases were included, one group given with combined QBSRDB therapy as treating one and the other with elemene injected into the thoracic cavity only as controlling one. Then, the therapeutic effects of different therapies were compared among these cases to see the clinical response to this medicine.From the experiments done as stated as above, the results obtained were shown as follows.1. Tumor-Inhibiting Effects of QBSRDB These experiments showed that the tumor-inhibiting rates for the animals with the cells of Lewis lung cancer transplanted were 28.38% ~35.44% and the survival duration-extending rates were 32.8%~41.41% for those with EAC cells transplanted respectively, when they were treated with QBSRDB. There were statistically significant differences present among them when compared with that of controlling groups (P<0.05).
