Part One The effect of angigenesis of magnesium lithospermate B in chick chorioallantoic membrane (CAM) modeObjective Danshen root has the long history of treat coronary artery disease.CAM is the effective model in studying angiogenesis. The effect of magnesium lithospermate B in CAM model is performed to test whether magnesium lithospermate B has the role of induce angiogenesis.Methods Chick chorioallantoic membrane (CAM) mode was made to study angiogenesis, saline was used as negative control and basic fibroblast growth factor was used as positive control, CAM vessels of different groups including several doses of magnesium lithospermate B were counted and compared to study whether magnesium litherspermate B could induce angiogenesis.Results The number of neo vessel of magnesium litherspermate B is significantly increased, compared with saline, but can't reach the bFGF level.Conclusions Magnesium litherspermate can strikingly induce angiogenesis in chick chorioallantoic membrane (CAM) mode.Part two The effect of magnesium lithospermate B on human endothelial cellObjectives The proliferation of endothelial cell is the important part of angiogenesis, but whether magnesium lithospermate B may stimulate the angiogenesis to induce the ischemia myocardium to create collateral circulation is not reported. The vitro study about the effect of magnesium lithospermate B on the proliferation of umbilical vein endothelial cell is performed to test whether magnesium lithospermate B could induce angiogenesis.Methods Effect of magnesium lithospermate B of different doses on proliferation ofhuman umbilical vein endothelial cells (HUVEC) was measured by MTT colorimetric assay and by tritium-thymidine-isotope assay, and cell cycle was measured by using flow cytometry respectively, saline was the negative control, and basic fibroblast growth factor was positive control, to test the role of magnesium lithospermate on the proliferation of HUVECs.Results: Magnesium lithospermate B could stimulate the proliferation of HUVECs in vitro and the similar change of morphology as basic fibroblast growth factor occurred.The cell proliferation is productive and the growth curve shifts to the supior quickly, and the absorbency of MTT is increased, and the adultaration of tritium-thymidine is improved.It hints that the magnesium lithospermate B might stimulate the cell proliferation. The role of magnesium lithospermate B has the correlation with its concentration. In the view of facilitate rate of cell proliferation, the rate of magnesium lithospermate B of low dose is 7.90%, the rate of middling dose is 71.07%, the rate of high dose is 2.27%, it is shown that low dose magnesium lithospermate B has the feeble facilitate effect. The effect is increased with the augment of dose until the high dose, the reason may be that osmotic pressure of high dose is increased to damage the cell membrane to inhibit the proliferation of the cell. The study of cell cycle showed that the ratio of G0 phase to G1 phase of the magnesium lithospermate B group is decreased and the ratio of S phase is increased, we can draw a conclusion that magnesium lithospermate B induces the cell of G0 from the quiescence state to G1 activation state and accelerates the cell of G, to S stage to synthesis DNA, and facilitates the proliferation of HUVECs.Conclusion: Magnesium lithospermate B can facilitate the proliferation of endothelial cell, the difference is significant compared with saline. It indicates that magnesium lithospermate B may induce angiogenesis.Part Three The effect of magnesium litherspermate B on the migration of human vascular endothelial cellObjectives Angiogenesis is a complicated process, at the mension of endothelial, it consists of several procedure such as migration, proliferation and tube-like structure formation.
