The Clinical And Experiment Study On Treatment Of Melasma And The Effect On Tyrosinase And Melanogenesis By Shen Zhe Qu Ban Granule

Object: In order to find out the curing and operating mechanisms, we afforded theoretical foundation by doaumentary research and compared the clinical effect of ShenZheQuBanGranule in YiSeZhiSeFa on Melasma. At the same time ,we studied the influence on the content of the SOD,E3,SP,the influence on tyrosinase activity and the proliferation and melanogenesis of B-16 murine melanoma cell, the influence on cavy models of Melasma.Method: 66 Cases of patients was divided randomly into the test group treated with ShenZheQuBanGranule and the control group with ShaJiGranule. The survey was conducted on clinical effect and the content of the E3,SP using radioimmunoassay(RIA), The external experiment of the influence on tyrosinase activity of ShenZheQuBanGranule by the rate of oxidation of DL-dopa and its effection to the proliferation and melanogenesis of B-16 murine melanoma cell by the method of cell culture in vitro. We also observed the changes of the number and morphology of melanocytes induced by UVB in the experimental Melasma model (brownish guinea pig).Results:1. ShenZheQuBanGranule had a good clinical effect, its whole effectiverate was better than that of ShaJiGranule(p<0.05)2. ShenZheQuBanGranule can reduce serum E3 like ShaJi- Granule(p>0.05).Its function of increasing SOD activity and reducing SP was also remarkable than that of ShaJiGranule(p<0.01) 3. ShenZheQuBanGranule can reduce the tyrosinase activity, which was better than that of ShaJiGranule(p<0.01) 4. ShenZheQuBanGranule can inhibit the proliferatio ,tyrosinase activity and Melanogenesis induced by UVB irradiation on B-16 Murine Melanoma, which was remarkable than that of ShaJiGranule(p<0.05)5. ShenZheQuBanGranule can inhibit the experimental hypermelanosis induced by UVB using brownish guinea pig model.Conclusion: ShenZheQuBanGranule, which was according to YiSeZhiSeFa, has the function of regulating the level of SOD,E3 and SP, inhibit the proliferatio ,tyrosinase activity and Melanogenesis. Its clinical effect remarkable than that of ShaJiGranule.
