Procyanidins are a class of polyphenolic compounds in plants. More and more researchers in the fields of nutrition, health and medicine have paid more attention to procyanidins owing to its potent antioxidants, anti-inflammatory activities, cardiovascular activities and so on. For a long time, the procyanidins of Grape seed(GSOPC) have been a focus research in the world. With the further studies, many scientists turned their research on other plant species in order to explore new procyanidins' resources.It is well known that the seedpod of Nelubo nucifera Gaertn. (Lotus Seedpod , LS) is a traditional medicine, which can remove blood stasis. But until now, people know little about the primary active components of this drug. The Lotus Seedpod procyanidins (LSPC) were firstly studied here by means of morden analysis technique, biotechnologies and pharmacological methods. The physicochemical properties, and its biological activities such as antioxidantion, cardiovascular activities were examined. And the possible mechanism of anti-myocardial ischemia was also discussed here. It was preliminarly proved that LSPC have the functions of antioxidantion activities and antinmyocardial ischemia, and that procyanidns were one of the primary active components of Lotus Seedpod for the first time. All of these studies provided a theoretical and experimental base for further research.1 Preparation and physicochemical property of LSPCFor the first time,the procyanidins content of non-edible parts of Nelumbo Nucifera Gaertn. was determined. The procyanidins extract of LSPC was obtained with Me2CO-H2O solvent system. Then it was isolated and purified by Sephadex LH-20 column chromatography, whose purity was more than 98 %.LSPC was a kind of buff powder and can produce cyanidins' reaction. It had good water solubility and better reducing ability than vitamin C. LSPC was stable inweak acidic solution, but unstable in alkaline solutions. The stability of LSPC could be significantly affected by Sunlight, temperature and metal ions.LSPC and OPC had maximum characteristic absorption only at 280nm in Ultraviolet spectrum, and their Infra-red spectrum were similar to Catechin. ES1-MS analysis showed that the molecular weight distribution of LSPC ranged from 291 to 1155, the main M+H peak values were 291.1, 579.2, 731.2, 867.2, 1019.4 and 1155.3 It also exhibited that LSPC contained monorners,dimmers and tetramers,in which the amounts of dimmers were more than others, and catechin and epicatechin were the base units in LSPC. 1HNMR and 13CNMR spectrums were confirmed that 2-4 polymers in LSPC were linked through C4-C8(or C6) bond .2 Antioxidative activities of LSPCEffect of LSPC on lipid auto-oxidization, lipoxygenase activies, free radical scavenging, erythrocyte auto-oxidization injury were studied here.The results showed that 0.1 % LSPC have strong antioxidant activity in soybean oil system, better than BHT with the same concentration. It was found that LSPC could inhibit lipoxygenase activity very significantly. The inhibitory rate of LSPC had gone beyond 90.0% with a concentration of 62.5 u g/ml, and the IC50 value was 21.6 u g/ml. In Fenton system LSPC didn't show pro-oxidant effect from 10 to 1000ug/ml,and the maximum inhibitory rate to OH was 90.1 %, and the IC50 value was 266.8 u g/ml; 31.2 u g/ml LSPC could effectively protect erythrocyte against auto-oxidation reduced by external factors, and the inhibitory rate reached 91.2%.3 Effects of LSPC on the function of hepatic antioxygenationThe effects of LSPC on the formation of MDA in liver homogenates incubated with Fe2+ and H2O2 in vitro was observed. Results showed: in vitro, LSPC could significantly inhibit the formation of MDA from liver homogenate and mitochondria, the inhibitory rate of LSPC with a concentration of 0.13 u g/ml could reach 92.3%.
