There are high prevelance of chronic atrophic gastritis (CAG) in China. The endoscopic examination rate of CAG ranged from 7.5% to 13.8% in general population, in which nearly 8% patients reported to develope into gastric carcinoma. According to the Correa's cascade of gastric carcinogenesis, gastric cancer was believed to develop from a multistep progression from CAG, intestinal metaplasia (IM), dysplasia (DYS) and subsequently to intestinal-type adenocarcinoma. In 1998 gastrointestinal pathologists reached a consensus on the definition of CAG, which was described with programmed loss of gastric gland and/ or replacement by intestinal glands in gastric mucosa. CAG was recognized close relationship with development with gastric cancer and listed as precancerous lesions in this meeting. Since CAG played an important role on the gastric carcinogenesis, so how to explore the development of CAG and reverse the patholigical change were critically essential.The reality of development of gastric carcinogenesis remains to further demonstration. The results was controversal and lack of convincing evidence in clinical research, which may induced by the bias of personal inherity of patients, specimen biopsy and the studying methods the authors referred to. So to establish the animal models, which could simulate the progression from CAG to gastric cancer, was definitely important. Animal model was available to assist in providing information to explore the progression of CAG, possible atrophic inducing factors, and develope new drugs. The imbalance in the cellular proliferation and apoptosis, and acquired multistep progress in molecular events could be attributed to the development from normal epithelial to atrophic gastritis. Animal model could investigate the molecular series of homegenous oncogene in human being and the possible regulating mechanisms in CAG.As we known, reversing intervention for CAG took an important role in the early prevention of gastric cancer. Eradication of Hp or long-term supplementation with antioxide agents, folic acid and p-carotene were approved to have benefit on reserving CAG, but still debated. It was lack of convincing strategy for reversal the pathological change of CAG. Our aims were to explore the effective drug on the reversing treatment for CAG.The experimental study on the development and reversal treatment of chronic atrophic gastritis and modern pharmaceutical exploiting with Muscovite included four parts, in order of "the establishment of CAG in experimental rats", "study of the regulating molecular mechanism of cell proliferation and apoptosis in rats with experimental CAG", "The effect of Muscovite on reversal treatment for CAG in rats" and "Modern pharmaceutical exploiting with Muscovite". The study was supported by the science development foundation of Zhejiang province and "5112" discovering program of Hangzhou city. We successfully established CAG models in rats and demonstrated the theory of CAG were induced by the damage of barrier in gastric mucosa and further destroyed in deep glandular tissues by inflammatory factors, and misbaiance in cell proliferation and apoptosis in gastric mucosa, subsequently, the gastric mucosa could not completely regenerate. The results shown that there were the change of expression of EGF/EGFR and other regulator peptide, high expression of oncogene, while low suppressor gene in CAG rat, which indicates of high trendy of carcinogenesis Muscovite could improve the restoration of mucosa barrier and glandular tissues, and accelerate with the reversing of atrophic lesions. Muscovite stimulated with endogenous secretion of EGF/EGFR, and inhibitored the expression of carcinogene C-erbB-2, as well as increasing the expression of tumor suppressor, which induced benign cell proliferation and normalized the turnover of cellular cycle.
