Study On Adenovirus Vector Based Vaccine Against Hantanvirus

Objective: To construct Hantaanvirus gene based Recombinant Adenovirus vaccine and to evaluate the immune response induced by the vaccine in animal models. Methods:1. The Hantaanvirus M fragment, Gl and G2 were amplified and cloned into T-vector for sequence analysis. The Gl and G2 genes were cloned into baculovirus vector for expression of the recombinant proteins respectively and identified the expression by Western Blotting assay and ELIS A.2. The recombinant adenovirus containing the Hantaanvirus M, Gl and G2 genes were constructed. The expression of the protein encoded by M, Gl and G2 was identified using Western blotting Assay.3. The BALB/c mice were intramuscularly immunized with the recombinant adenovirus alone or combined with the DNA vaccine. The specific immune response including humoral and cell-mediated immunity and mucosal immunity were analyzed. The protective effect of the recombinant adenovirus was evaluated by challenging the immunized hamsters with the vaccinia virus containing the HTNV M fragment.Results:1. The recombinant Gl and G2 proteins were expressed successfully in insect cells and the proteins reacted specifically with the HFRS patient sera and positive rabbit serum.2. The Hantaanvirus-gene based adenovirus vaccine induced specific immune response in mice and hamsters. The vAd-M elicited higher level of neutralizing antibodies and stronger cell-mediated immune response than those of the vAd-Gl. The vAd-M vaccine could protect hamsters from infection with recombinant vaccinia virus expressing Hantaanvirus M fragment.Conclusion: Hantaanvirus M gene based recombinant adenovirus vaccine can induce specific immunity against Hantaanvirus infection. The Adenovirus based vaccine might be an alternative promising candidate vaccine against hantaanvirus.
