The Effect Of Low Frequency, Low Energy, Pullsing Electromagnetic Fields (PEMFs) On Neoadjuvant Chemotherapy And Metastasis Of Osteosarcoma Cell Line

[Purpose] I will investigate the action of anticancer drug on HMEC-1cell under PEMF stimuli. [Method] The HMEC-1 cells were cultured in 25cm2 culture flask in RPMI 1640 medium with 1% HEPES buffer and 10% FBS at 37℃ in a incubator with 5%CO2 . Cultured cells in the logarithmic multiplication period, suspended with the culture medium containing or not containing VEGF (15ng/ml) at 1.5×10 4 cells /ml , then 200μl of it were inoculated in each well of 96-micro well plate for 48 hours. Doxorubicin and Cisplatinum were adjusted to 6 concentration gradients as 0.5,1.0,1.5,2.0,2.5μg/ml, which were applied on culture medium with the same concentration Then one group was exposed to PEMF stimulation for 12 hours after inoculation and continued incubation for 24 hours, another group without the PEMF stimulation were cultured in another incubator for the same hours as the control. Frequency 200Hz, the pulse width 100μsec, magnetic field strength 50 Gauss, stimuli time 12 hours were designated as the optimum condition of PEMFs. After this period, cell number was counted with WST-1 method. [Result] 1, Cell growth by VEGF: The number of HMEC-1 cells were increased by VEGF. 15ng/ml of VEGF showed maximum effect with statistically significant. 2, Anticancer drug free group: The number of HMEC-1 cells were slightly increased by PEMF stimulation, however, statistically significant change was not observed in neither group in which VEGF was added or not. 3, Anticancer drug group: ①VEGF free group: The number of cells were decreased depending on the concentration of anticancer drug. At low concentration of DOX and CDDP, no more than 1.5ug/ml, did not show significant difference under PEMF stimuli. At high concentration of DOX and CDDP, more than 1.5ug/ml furthermore, the number of cells were more decreased with significant difference between PEMF (+) group and PEMF (-) group.② VEGF group: The number of cells were decreased significantly even with low concentration of DOX and CDDP, more than 0.5ug/ml, between PEMF(+) group and PEMF(-) group. However the number of cells did not show significant difference at concentration above 2.0ug/ml in both group. [Conclusion] The effect of anticancer drugs on the HMEC-1 cell was enhanced by PEMF stimuli. Activated HMEC-1 cell with VEGF showed much more sensitively to anticancer drugs and the number of cells were significant decreased even at low concentration of anticancer drugs.
