Objective Reportedly, increased serum phospholipase A2(PLA2) activityand its damaging effects on alveolar type II pneumocytes are associated with the pathogenesis of lung injury in acute hemorrhagic necrotizing pancreatitis(AHNP). Rat alveolar type II pneumocytes were isolated and cultured. The damaging effects of PLA2 and AHNP rat serum on alveolar type II pneumocytes were examined in vitro.Methods Rat alveolar type II pneumocytes were isolated and cultured and were stimulated with PLA2 (50ng/ml)and AHNP rat serum, with or without administration of the PLA2 inhibitor-quinacrine. Four hours later, the lactic dehydrogenase (LDH) level of the supernant and NF- k B activation of cultured pneumocytes were detected. In addition, the ultrastructural changes of the cultured pneumocytes were examined by transmisson electron microscopy. Results In vitro, the serum with high PLA2 activity and PLA2 (50ng/ml) induced NF- k B activation by alveolar type II pneumocytes and induced significant amounts of LDH production and cytotoxicity toward type II pneumocytes examined by transmisson electron microscopy. These activities were significantly inhibited by quinacrine. Conclusions PLAt causes alveolar type II pneumocytes injury in AHNP and this injury is associated with NF- k B activation induced by PLA2 besides its direct injury effects on type II pneumocytes.
