In people, cardial arrhythmia is always accompanied by cardiovascluar system disease,such as viral myocarditis, cardiomyopathy, hypertension and heart failure. Every year, halfsufferers died from this kind of serious heart disease, which is difined to Cardiac SuddenDeath by World Health Organization. This kind of sudden death is usually causing byventricular fibrillation led by myocardial ischemia. A statistic shows that 95% arrhythmiahas direct relationship with stress. Now, stress has been comfirmed to be one importantinducement of cardiovascluar system disease. Antiarrhythmic drugs usually induce proarrhythmia and some other toxic and sidedffects . The combined use of Tradditional Chinese medicine could produce a variety ofcomplicated changes , such as increase of drugs efficacies, elimunation of toxic and sideeffects. But until now there are no effective components to be reported and besides, nospecial drugs to be used for arrhythmias induced by stress. Our subject for reseach is a famous traddional chinese formula-Fumaitang (FMT). It iscomposed of Radix Glyzyrrhizae preparata, Radix Ginseng, Radix Oplnopogonis and etc. Itcan benefit Qi, nourishing the blood and Yin, restore pulse beating and be also indicated forQi and blood deficiency, palpitation, dry throat and tongue, and feeble and rapid pulse andetc. FMT, compared with ZhiGanCaotang, has less composer and more explicit effectivecomponents. According thus, so we comsider FMT as a remarkablely valuable subject tostudy. This reseach was concentrated on the effects of FMT and its effective components ontriggered activity, triggered arrhythmia, and myocardial damage induced by stress. Theexperment results are as follows.1. The study of phytochemistry In our premare clinical practice, we found that FMT has very perfect curative effect toventricular premature (VP) and artia premature. However, tradditional water decction couldbring large foreign matters, so we have done lots experiments to study the extract methodand technology on FMT besides, its preparation technology had been studied either.1.1 Study of Fumai Decoction extract methods: comparing Maceration with Decoction and Percolationunder the same conditions as possible. The content of glycyrrhizinic acid and ginsenoside Rb1 acquiredwith previous extract methods were determined by HPLC. The results showed the content of glycyrrhizinic acid and ginsenoside Rb1acquired with Maceration was higher than other methods and impurity was also less. So Maceration wasselected to be the extract method of Fumai decoction.1.2 Study of Fumai decoc tion extract technic: uniform design test was used to optimize Macerationcondition, ethanol concentration, ethanol volume, extracting temperature and time were observed, fourelements three levels, U9(94) Uniform table was used, and the content of glycyrrhizinic acid andginsenoside Rb1 was determined by HPLC. The results showed that optimal extraction condition was asfollows: the raw drugs were extracted by 10 times of 50% ethanol at 60℃ for 4 h.1.3 Study of effective part in Fumai decoction: Fumai decoction crude extract was purificated withmacroporous adsorption resin, the effect of ethanol concentration on the content glycyrrhizinic acid andginsenoside Rb1 was observed with the same elution rate (1mL·min-1) and the same elusion volume (6times of resign bed volume). Glycyrrhi-zinic acid and ginsenoside Rb1 were considered as effective partin Fumai Decoction, the content of glycyrrhizinic acid in different concentration ethanol elusion wasmeasured by HPLC and the content of ginsenoside Rb1 was determined by Spectrophotography.
