Experimental Research To Confirm The Effective Components In SQ Complex Prescriptions Treating The Uterine Bleeding Of Cu-IUD-Wearing Rabbits

In order to confirm the effective components in SQ complex prescriptions which was composed of Radix notoginseng and Radix rubiae, an animal model of Cu-IUD-wearing rabbits was established and 6-K-PGF1, TXB2, t-PA, hemorheology markers, endometrium micro vessel count were used as the screening markers. Several animal tests were taken. The results showed :l.The animal model of rabbits with Cu-IUD-wearing had pathological alteration with manifestates of biochemical marker alterations, hemorrheologic abnormality, endometrium hyperemia etc. SQ complex prescriptions had therapeutical effects, could ameliorate these abnormal alterations.2.Radix notoginseng and Radix rubiae had therapeutical effects when apply alone, but their effects had not as good as combinational apply.3. The chief effective components of Radix notoginseng were Panax notoginseng saponin (PNS) and dencichine. The effective component of Radix rubiae was mollugin. All of these three effective components could ameliorate abnormal hemorheology alterations and biochemical marker alterations. The rest extract of Radix notoginseng and Radix rubiae had fewertherapeutical effects.4. PNS, dencichine and mollugin all had significant effects on ameliorate abnormal alterations. The best prescription matching was PNS 240 mg, dencichine 24 mg, mollugin 100 mg (one day)The extration process of PNS, dencichine and mollugin were established and optimized by several tests. The results showed :1 .Mollugin was extracted with ethanol from Radix rubiae by heating reflux and purified with DIOI macroporous resin, its content up to 70%.2.PNS was extracted with ethanol from Radix notoginseng by infiltrating, and purified with D101 macroporous resin, its content up to 90%.3.The coarse dencichine was isolate from the total ammo acid with CM Sephadex C - 25, the later was isolate from the Radix notoginseng solution with 732 cation exchange resin. The coarse dencichine could apparent shorten hemostasia time by cutting mice caudal vein tests.
