In recent years, with the development of social economy and the improvement of people's living standard, there has been an increse in the incidence rate of hyperlipemia, diabetes mellitus, obesity, and other diseases. Under the above lipodystrophic pathological condition, the body is liable to complicate with nonalcoholic fatty liver diseases (NAFLD), such as nonalcoholic fatty liver, nonalcoholic steatohepatitis (NASH), etc. NAFLD represents a spectrum of disorders characterized by predominantly macrovesicular hepatic steatosis that occur in individuals even in the absence of consumption of alcohol in amounts considered harmful to the liver. For over 20years, Our Institute of Hepatopatry has been engaged in the research on the effect and mechanism of IETM on the development and progress of various liver diseases .By activating kupffer cell (KC), IETM plays its important role in causing various acute and chronic hepatic disease to become chronic and severe and even hepatic failure . So far, there has been no reports on the study whether the body in NAFLD is accompanied by IETM , what effect IETM has on NAFLD, especially on the development of fatty liver into NASH. In order to demonstrate the effect of IETM on NAFLD and its mechanism, and meanawhile to observe the effect and mechanism of glycine on rat models with NAFLD to provide a theroretical basis for the lack of effective drugs in the prevention and treatment of NAFLD.The experiment was carried out in four parts,Chapter1. Study on the copy of NAFLD modelsChapter2. Study on the effect and mechanism of IETM caused by corn oil and the formation of NAFLD Chapter3. Study on the changes of phagocyted funciton of KC in the formation of NASH Chapter4. Study on glycine in antagonizing IETM in NAFLD Chapter 1. Study on the copy of NAFLD modelsExperiment 1 The role of corn oil in the formation of NASH The results of the experiment showed that, using 20% corn oil to feed the animals for 9 weeks, serum and Liver Lipid Levels were increased, and plasma ET and ALT contents elevated. Liver Showed the pathological changes of steatohepatitis on histological examination.Experiment 2 The role of corn oil in the formation of nonalcoholic fatty Liver fibrosis The results indicated that using 20% corn oil to feed the animals for 12 weeks , TG level of liver was increased, and ET and ALT contents of plasma elevated, on histological examination, it was found that inflammatory cell infiltration and spotty necrosis of Liver cells were aggravated on the basis of the changes of steatohepatitis. Fasciculate collagenus fibers distributed along vascular ware seen on VG staining . Hepatic pathology diagnosed them as mild and moderate steatohepatitis, and focal perisinous fibrosis developed in liver acinus's area 3 zone.Experiment 3 The role of saturated fatty acid and unsaturated fatty acid in the formation of NASH in rats Compound agents , dominated by carbon tetrachloride, to which saturated fatty acid or unsaturated fatty acid were added, were given the animals for 3 weeks. The results showed that their weight was reduced and hypoevolutism was present. Liver lipid was aggregated, and the contents of plasma ET, ALT, MDA, TNF-α, TGF-β1 and the contents of HA, LN and liver collagen were higher in the experimental rats than in the control rats. The changes of NASH occurred on Liver histological examination .However ,The contents of plasma ET and TNF-α were significantly higher in unsaturated fatty acid group than those in saturated fatty acid group, and the contents of HA , LN, TGF-β1 and Liver collagen were also significantly higher in unsaturated fatty acid group than those in saturated fatty acid group on histological examination, relatively remarkable fatty infiltration was found, and cystic fatty degeneration Liver cells fused into was also seen. unsaturated fatty acid group showed remarkable hyperplastic collagenus fibers compared with saturated fatty acid group . on transmission electron microscope (TEM), fat-storing cells appeared sat...
