Periodontal tissue regenerating in tooth movement has been regulated by nerve system. It is known that SP and CGRP play important roles in periodontal tissues,but deeply research is scarcity. In this study, molecule biology technique, immunohistochemical methods ,cell culture and inspect technique had been used to investigate the relationship between CGRP and periodontal tissues regenerating in rats tooth movement, and discuss the effects and mechanism of CGRP in tissues and cells. These are the main study contents and results : 1. Changes of Periodontal tissue during and after tooth movement in ratsIn this experiment, HE colorate method has been used to study the periodontal tissue remodelling when give force to move the first molar of rats and stop to move the first maxillary molar, set 11 groups of rats:compared group, given force after 2 hour,4 hour, 8 hour,lday, 3 day, 7 day, 14 day, and unloading the force after 7 day, 14 day, 28 day. We observed that on the strain side, space of periodontal ligament increased, blood vesselsdilatated, osteoblast appeard on alveolar bone, and alveolar hyperplasia. On the press side, the space of periodontal ligament decreased, osteoclast appeard, and the alveolar bone has been absorbed. After stop given force on the first molar, periodontal tissue resumed gradually. This results illuminate the process of the periodontal tissue regeneration in and after tooth movement.2. Expression of CGRP1 receptor on dental hard tissues in peridental tissue in ratsIn this study, the method of hybridization in situ were used to investigate the existence and localization of CGRP1 receptor in rat dental pulp and hard tissues and in peridental tissue. This results showed that CGRP1 receptors were distributed in the endothelial cells of the larger blood vessels in dental pulp and peridental tissue, did not found CGRP1 receptors in dental hard tissues. Combined this results to other researches, we got conclusion that The CGRP1 receptor were distributed in the larger blood vessel and through CGRP1 receptor, CGRP have effects on larger blood vessel, regulate blood stream and transfer signal of cells.3. Change and meaning of CGRP-IR nerve fibre in rats dental pulp and peridental tissue during and after tooth movement.The present study was designed to investigated the changes of CGRP innervation during and after tooth movement of first maxillary molar in the rat. It has the same grouping as experimentone. In the result, the amount of CGRP-IR nerves in rat first maxillary molar pulp were acutely decreased after tooth movement, then come back gradually, and catch the most quantity after 24 hours. CGRP-IR nerves in peridental tissue have similar change. It quasi the status which from acute trauma to chronic trauma. When the tooth were stimulated, nerve fibres released neuropeptide, sent afferent signal to nerve centre and joint local inflame response.It indicates that CGRP play an important role in periodontal tissue remodelling in tooth movement.4. Expression of CGRP in trigeminal ganglion during and after tooth movement in ratsIn this study, RT-PCR methods were used to research the changes of CGRP contents in trigeminal ganglion during and after move rat first maxillary molar, and still has the same grouping as experiment one. It discovered that CGRP in trigeminal ganglion decrease firstly, then increase fast and more high than compare group, and decrease again after three days. This result shows that trigeminal ganglion released neuropeptide to nerve fibres when get the afferent signal from sense nerves firstly, and synthesize neuropeptide thereafter. Tooth movement caused periodontal tissue remodelling,not only local neuropeptide,but also nerve centre have been concerned with this process, regulate the periodontal tissue regenerating together.5.
