AIM: This research used many modern scientific technology, from the whole-cell-protein-gene etc different levels, to observe the influence of Shenmai injection on heart function and ventricular remodeling after acute myocardial infarction, and discuss the interference effect of Shenmai injection on myocardial hypertrophy and apoptosis that is two important factors in ventricular remodeling pathologic process. The experiment included two parts: In vivo research: the effect of Shenmai injection in improving heart function and ventricular remodeling on acute myocardial infarction rat In vito research: discuss the interference effect of Shenmai injection on myocardial hypertrophy and apoptosis Part one: the effect of Shenmai injection in improving heart function and ventricular remodeling on acute myocardial infarction rat 1. Method: Seventy six healthy male SD rats born 10-12 weeks were ligated left main coronary artery to make the model of acute myocardial infarction, then randomly divided into six groups. â‘ MI 1 week group: model group after myocardial infarction 1 week,â‘¡SM1 weeks group: treated with Shenmai injection 1 week after myocardial infarction, â‘¢MI 2 weeks group: model group after myocardial infarction 2 weekâ‘£SM2 weeks group: treated with Shenmai injection 2 weeks after myocardial infarction, and two sham-operate groups .Every group was administered Shenmai injection by i.p. from the day the model was made till drawing materials. The dose of Shenmai injection was five times of normal dose of human. Model group and sham group were administered the same dose NS by i.p. Using polygraphy measure and analyse heart function parameter: mean artery pressure, left ventricle systolic pressure, left ventricle end diastolic pressure, left ventricle +dp/dtmax and -dp/dtmax, reflected the function of systole and diastole. Disposing blood and cardiac muscle respectively, measuring heart weight and left ventricle section diameter, and calculating ventricle weight index; determining the content of Angâ…¡and ANP in blood and heart muscle by radioimmunoassay; observing tissue structure changes by HE dyeing, and Masson dyeing; observing ultrastructure changes by electron-microscope. 2. Result(1)After myocardial infarction caused by coronary artery ligating, LVSP, +dp/dtmax and -dp/dtmax decreased, LVEDP increased. Shenmai injection could improve these changes obviously. It indicated that Shenmai injection could improve left ventricle systole and diastole function after acute myocardial infarction. (2)Treated with Shenmai injection, it could alleviate cardiomyocyte degeneration in infarcted region; myofibril and intercalated disk were more regularly than the model group. Mitochondria swelled, crista lightly ruptured even vacuolization, but it was more alleviated than model group in corresponding time. Nucleolus chromatin structure of cardiomyocytes was close to normal; sarcoplasmic reticulum was lightly dilated, filament arranged in order, no obviously necrosis; interstium swelling was not distinct; collagen hyperplasia lessened, few inflammatory cells could be observed. (3)Compared with sham-operate group, heart wet weight, ventricle weight index and left ventricle section diameter notably increased after myocardial infarction caused by left coronary artery ligating. Shenmai injection could distinctly improve these changes. Model group of one week had increased, but it had no statistics significance. The collagen content of infarcted region and non-infarcted region increased with time after MI. (4)Shenmai injection could significantly reduce Angâ…¡and ANP content of blood and heart muscle after acute myocardial infarction, especially the ANP of circulation.
