Clinical meta analyses approve of that transfusion induces recipients immunosuppression so that may increase incidence of bacterial infection tumor recurrence. We started with APC to study T cells function. If APC presentation function is handicaped, the whole T cells proliferation will be restrained. While raising the APC number, T cells proliferation will be enlarged. Using anti-CD3 and anti-CD28 to mimic stimulatory signal I / II, to study their function of activing T cells in order to explore the possible mechanism of immune up and down regulation. Basing on that we may find the key to know how transfusion make side effects and how to correct them.1. Study on FHb, PRBCs and leukoreduced PRBCs' suppressing two-way MLRUsing two-way MLR, cells culture and incorporation of 3h-thymidine (3H-TdR), we added PRBCs, etc at different ratio to reaction system, then raise the concentration of APC with normal APC concentration MLR as negative control and CsA as positive control. Calculated the suppressive ratio of above factors to MLR. Along with the rise of added PRBCs volume, its suppressive effect grew. When removing the leukocytes of PRBCs may reduce that effect. FHb can impact MLR. When the culture system contain twice APC number, the suppressive effect of PRBCs become weaker. RBC and other factors may disturb the APC function to suppress MLR. We may use this method to study or prove the immunomodulation of large volume transfusion and leukocytes removing.2. Effect of anti-CD3/28 on enhancing lymphocyte transform function Study the T cells function with the methods of incorporation of 3h-thymidine (~3H-TdR) for measure lymphocyte transform function LTF, cell culture and flow cytometry (FCM), using coated anti-CD3 and anti-CD28 as stimulator.
