Effect Of Bone Marrow Mesenchymal Stem Cells Transplantation On Aortic Remodeling And Target-organ Protection In Isolated Systolic Hypertension Rats

Objective To explore the structural and functional changes of aortic wall, heart and kidney in isolated systolic hypertension rats and SHR after MSCs transplantation and make clear the effect of MSCs on aortic compliance and organ-damage repair so as to provide new idea for hypertension therapy, especially for isolated systolic hypertension.Methods Get mesenchymal stem cells(MSCs) form health Wister rat and establish a steady culture system in vitro. Carry out the purification , identify and marking in vitro. ISH model rat by warfain and SHR were selected to receive MSCs infusion respectively, 4 weeks later, PWV, blood pressure and cardie pressure-volume curve were measured at the same time. Elatic/collagen ratio, type I /III collagen , calcification degree were measured according to different coloration methods. The expression of hemoxygenase,fibronectin carried out by immunochemistry. Osteopontin were explored by western-bloting. Calcilium density were canculated in each group with atom-spectrum.Results 1.The effects on aortic: MSCs infusion can't change the trends of systoleic blood pressure in all hypertensive rat, but it can do reduce elastic/collagen ratio and pulse wave velocity(178.6526±18.5494)in ISH rat compared with the control group(P<0.01). The expression of FN and HO were increased and OPN was decrease compared with control group(P<0.05). By correlation analysis we found PP change may correlation with PWV variety.
