The purpose of our studies is try to gain a better insight into the role of Rosiglitazone on high glucose-induced NF- κ B activation in MCP-1 and ICAM-1 by mesangial cells. The growth inhibiting effect of Rosiglitazone on mesangial cells was measured by MTT and CCK-8 methods,The levels of MCP-1 and ICAM-1 in the supernatants were determined by enzyme-linked immunosorbant assay (ELISA.).MCP-1 and ICAM-1 mRNA expression were measured by semi-quantitative RT-PCR assay. Activation of nuclear factor- κ B(NF- κ B) of rat mesangial cells were measured by electrophoretic mobility shift assay (EMSA). Our studies suggest that hyperglycemia can stimulate mesangial cells proliferation in 48 hours. Rosiglitazone,in the range of concentrstions from 10~20 μ mol/L, has a capability of inhibiting proliferation in high glucose cultured mesangial cells. Rosiglitazone can inhibit high glucose-induced MCP-1 and ICAM-1 mRNA and protein expressions in mesangial cells.In high glucose condition,the activation of NF-κ B is increased,and Rosiglitazone can partly block high glucose induced activation of NF- k B. These findings may provide an experimental evidence for further evaluating the possibly protective effect of Rosiglitazone against diabetic nephropathy.
