First partThe Influence on MSCs' proliferation and morphology by the Microgravity Rotating SystemObjective: Investigate mesenchymal stem cells(MSCs) proliferation changing after which were cultivated under microgravity rotating culture system. Methods: MSCs were obtained from about 3ml bone marrow which was aspirated from adult New Zealand Rabbit, and were purified with adhering and medium changing. L-DMED+20%FBS was used as culture medium. MSCs obtained were cultured in vitro and passed several times. The change of morphology was observed during the passage cultivation of MSCs. Immunohistochemistry was used to detect the expression of CD71and CD44. After combined with PGA and chitosan, MSCs were cultured under the microgravity rotating cell culture system(RCCS).The proliferation of MSCs was evaluated by MTT assay, and the biological features of the compound was also observed under the scanning electron microscope.Results: Cultured under the RCCS, the proliferation of MSCs was enhanced markedly. And the amelioration of the MSCs'biological features was also found by the canning electron microscope.Conclusion: The RCCS can greatly improve the proliferation of the MSCs, and ameliorate the biological features of the MSCs.Second partThe Influence on the Chondrogenic Ability of the MSCs in vitro by the Micorgravity Rotating Culture SystemObject: Investigate the influence on the chondrogenic ability of the MSCs in vitro bymicrogravity rotating culture system.Methods: After combined with the three dimensional carrier— chitosan, MSCs ofgeneration 3rd were divided into two groups. One was cultured in the microgravity rotating culture system, the other was cultured in the static culture system as usual.
