many reseach had reported that ginsenoside Rg3 have the ability to inhibited the tumor invasion and metastasis,and The Ley antigen is a blood group-related difucosylated oligosaccharide The Ley antigen has been shown to be expressed by 60–90% of human carcinomas of epithelial cellorigin, including breast, pancreas, ovary, colon, gastric, and lung cancer and it si related to the metastasis,we use the breast cancer cell MCF-7 to investigated the relationship between the metastasis and ley,furthermore to study the mechanism of ginsenoside Rg3 to inhibited the cancer metastasis,what we search on blow. 1.Effect of gensenoside Rg3 on the expression of LeY and fucosyltrans-ferases (FUT4) in breast cancer MCF-7 cell line To investigate the impact of ginsenoside Rg3 on the expression of Ley oligosaccharide in breast cancer cells (MCF-7) and fucosyltransferases (FUT4). Method The effect on the expression of Ley oligosaccharide upon usage of Rg3 was determined by Immunochemical staining, immunofluorescence, flow cytometic analysis and Western-blot. The effect on the expression of (FUT1,4) upon the usage of ginsenoside Rg3 was determined by RT-PCR. Results: After use of ginsenoside Rg3, Immunochemical staining and immunofluorescence revealed a fading in Ley oligosaccharide. Western-blot revealed a significant decline in LeY level (p<0.05), RT-PCR revealed a decline in the expression of FUT4, correlating with the decline in the expression of Ley oligosaccharide. Conclusion: ginsenoside Rg3 can effectively suppress the expression of Ley oligosaccharide and fucosyltransferases (FUT4) 2.Inhibitory effect of gensenoside Rg3 on the expression of Matrix Metalloproteinases in breast cancer MCF-7 cell line Objective To investigate the effects of gensenoside Rg3 on the expression of matrix metalloproteinases (MMP) in human breast cancer MCF-7 cell line Methods The expression and activity of MMP of MCF-7 cells with and without treatment of genesnoside Rg3 were measured by Immunohisto-chemistry, gelatin zymogram respectively1 Results ①Treatment with increasing dose of gensenoside Rg3 in MCF-7 cells caused a dose dependent decrease in the expression of MMP2 and MMP9 on MCF-7 cell ( P < 0.01, respectively)②the secretion of MMP2 and MMP9 on MCF-7 cell is reduce by the increasing dose of gensenoside Rg3 ( P < 0.01 ,respectively) Conclusion the result show that the inhibit invasion mechanism of gensendside Rg3 is decrease the secret of matrix metalloproteinases MMP-2,MMP-9. 3.Effect of gensenoside Rg3 on the DAG/PKC signaling pathways in breast cancer MCF-7 cell line The growth and metastasis of tumor cells are ssociated with DAG/PKC signaling pathways. In order to understand the mechanism of the ginsenosede Rg3 to effect on the PKC signaling pathways,the translocation of PKC isoforms in MCF-7 cell and the content of dia-cyclglycerol was compared between cells treated with ginsenosede Rg3 and control cells.
