Objective:1. To check and measure EZH2 gene mRNA expression in ovarian cancer cell line.2. To study the the relationship between ovarian cancer and EZH2 gene mRNA. Methods:1. To testify the expression of EZH2 gene, we applied reverse transcription -polymerase chain reaction (RT-PCR) to check and measure EZH2 gene mRNA in ovarian cancer cell line SKOV—3, and measure the sequence of RT —PCR's production.2. We applied RT—PCR to check and measure normal ovarian epithelial tissue and ovarian epithelial cancer tissue and confirm the rates of expression of EZH2 gene mRNA and the level of EZH2 gene mRNA in normal ovarian epithelial tissue and ovarian epithelial cancer tissue.Results:1. There really have the expression of EZH2 gene mRNA in PC3 and SKOV-3 cell lines.2. There are 3 cases' expression of EZH2 mRNA in 10 normal ovarian epithelial samples (30 %), and There are 24 cases' expression of EZH2 mRNA in 28 in ovarian epithelial cancer samples (85.7 %), there is significant difference between the rates of expression of EZH2 mRNA in normal ovarian epithelial samples and ovarian epithelial cancer samples by statistical analysis ( P < 0.05). There are 13 cases' expression of EZH2 mRNA in 16 serous cystadenocarcinoma samples, and the level of expression of EZH2 mRNA is 1.46+0.44. all 10 mucinous cystadenocarcinoma samples have the expression of EZH2 mRNA, and the level of expression of EZH2 mRNA is 1.52+0.48. there is 1 case's expression of EZH2 mRNA in 2 ovarian endometriod cancer samples, there is no significant difference between the levels of EZH2 mRNA's expression in serous cystadenocarcinoma samples and mucinous cystadenocarcinoma samples by statistical analysis (P>0.05) .There are 13 case's expression of EZH2 mRNA in 15 cases low differential ovarian epithelial cancer samples, and the level of expression of EZH2 mRNA is 1.55+0.43. there are 10 case's expression of EZH2 mRNA in 11 intermediate differentiation samples, and the level of expression of EZH2 mRNA is 1.41+0.50. there are 1 case's expression of EZH2 mRNA in 2 high differentiation samples, there is no significant difference between the levels of expression of EZH2 mRNA in low differential ovarian epithelial cancer samples and that in intermediate ovarian epithelial cancer samples by statistical analysis (P>0.05) . There are 7 cases (1.66+0.33)which level of CA125 is not more than 35mIU/L and 17 cases (1.42 + 0.47)which level of CA125 is not less than 35mIU/L in 24 cases'expression EZH2 mRNA of ovarian cancer samples. There are 13 cases(1.42± 0.46) which level of CA125 is not more than 200 mlU/L and 11 cases (1.57 + 0.43)which level of CA125 is not less than 200 mlU/L in 24 cases' expression EZH2 mRNA of ovarian cancer samples, there is no significant difference between the levels of expression of EZH2 mRNA 13 cases which level of CA125 is not more than 200 mlU/L and 11 caseswhich level of CA125 is not less than 200 mlU/L in 24 cases' expression EZH2 mRNA of ovarian cancer samples by statistical analysis (P>0.05) . There are 13 cases(1.43±0.45) who are not more than 50 years old and 11 cases(l.62+0.35) who are not less than 50 years old in 24 cases' expression EZH2 mRNA of ovarian cancer samples, there is no significant difference between the levels of expression of EZH2 mRNA in 13 cases who are not more than 50 years old and 11 cases who are not less than 50 years old in 24 cases' expression EZH2 mRNA of ovarian cancer samples by statistical analysis (P>0.05) . Conclusions:The experiment is the first study of relationship between EZH2 gene mRNA and ovarian epithelial cancer. Because the number of ovarian cancer cases is finite, the result of experiment is waiting to magnify the number of ovarian cancer cases to validate. The relationship between EZH2 gene and other genes which have any relationship ovarian epithelial cancer await to be studied.
