Objectives: The induction of differentiation therapy for cancer is a new field in the research of tumor treatment. Nowadays, there are many related materials reported the research of non-solid tumors. The difficult problem is how to induce effectively differentiation on the solid tumors for a long time. The aims of this study is to probe the effects of induced differentiation by auxin on oral squamous cell carcinomas and search the rational inducing materials. Moreover, the mechanism of induced differentiation was explored, the experimental gist and theoretic element were offered for the induced differentiation for oral squamous cell carcinoma, and the therapy of oral squamous cell carcinoma was developed and improved.Methods: The changes of surface differentiation markers, cell configuration, restrain of cell growth, ultrastructure, the expression of Caspase-3 mRNA, cell apoptosis, content of DNA and the ability of suppressing tumor growth were examined by using light microscope(LM), transmission electron microscope(TEM), flow cytometry(FCM), immunohistochemestry (IHC), in situ hybridization and TUNEL method through establishing a model of OSCC-cell-inducing differentiation in vitro and in invo.Results: In our study the restraint of cell growth were controlled by ABA, JA and MJ. There was a trend that the tumor cell had transformed the normal cell. Furthermore, the time-dosage dependent relationship existed in the inhibition rate of tumor cells. The results showed that the expressions of involucrin protein and retinoic acid receptor p in experimental group had been higher that of control group so as to upraise the level of caspase-3 and start up the cell apoptosis. The DNA index of tumor cell declined. The proportion of Go/Gi anddM phase rised distinctively while that of S phase went down. Moreover the apoptotic index of tumor cell went up in experimental group. Among ABA, JA and MJ the ability of inducing differentiation was MJ, JA, ABA from mightiness to feebleness in turn.Conclusion: hi vitro and in invo the obvious inhibition of tumor cell proliferation was caused by auxin (ABA, JA and MJ) which had induced the tumor cells to transform the mature cell or immediately mature cell and furbished the prototype and function of normal cell so as to result in cell apoptosis. These auxins are regarded as the material of induced differentiation on oral squamous cell carcinoma. The possible mechanism is that auxin acted on the tumor cell and ascended the level of RAR(3 gene through combining the correlative receptors so that increased the expression of involucrin protein, transformed the tumor cell to the mature cell or immediately mature cell, promoted the activity of caspase-3 and resulted in apoptosis of tumor cell.
