Objective: To observe the anti-angiogenetic effect of interferon alpha on the human liver cancer cell strain smmc-7721 nude mice model, and investigate the mechanism of it related to the vascular endothelial growth factor (VEGF). Thus offer information for the clinical use of interferon alpha to treat tumor angiogenesis.Methods: The smmc-7721 cell strain was amplified and then injected to nude mice subcutaneously to establish the nude mice model. A certain amount of interferon alpha was given to the experimental group according to the weight of nude mice for every another day, while giving only the same amount of saline to the control group. After 15 times, kill all the nude mice. We then measured the diameter of each tumor tissue. We also compare the difference between the two groups using the methods below: VEGF immunohistochemical quantitative analysis; VEGF mRNA in situ hybridizational quantitative analysis and microvessel density (MVD),andfinally analyze all the results to elucidate the mechanism of interferon alpha on tumor anti-angiogenesis.Results: The average diameter of tumor tissue in the experimental group was 0.74 + 0.30cm, while that in the control group was 1.37 + 0.27cm. Their difference was statistically significant ((PO.01); hi VEGF immunohistochemical quantitative analysis, the average IOD value in the experimental group was 5.81+2.70, while that in the control group was 10.83 + 5.24, statistically showing that the experimental group were lower than the control group; In VEGF mRNA in situ hybridizational quantitative analysis, the IOD value in the experimental group was 6.61+2.82, while in the control group it was 10.39 + 3.29, there had statistical significance between the two groups; And finally compare the MVD between the two groups, in experimental group it was7.41 + 1.63, while in control group it was 10.14+1.82, showing statistically difference.Conclusions: Interferon alpha can suppress the angiogenesis of tumors on the nude mice model of the human liver cancer cell strain smmc-7721. The mechanism is that it can suppress the expression of VEGF in tumors.
