Objective:To investigate the role of Cyr61 in the pathogenessis and development of autosomal dominant polycystic kidney disease(ADPKD). Methods: Cyr61 protein quantity in ADPKD cyst-lin-ing epithelial cells and body fluid were detected by Western blotting, and its localization wasexamined by immunofluorescence and in situ hybridization.A recombinant plasmid pcDNA3.1+Cyr61 was constructed and transfected into HKC.Then, the component of ECM , the proliferation rate, apoptosis exponent and some proteins related to proliferation and apoptosis were detected. Two hairpin siRNA template DNA of Cyr61 were synthesized and ligated into pSilencer 2.1, then the siCyr61 plasmid with the better suppression was selected and transformed into cyst-lining epithelial cells. Then, the component of ECM , the proliferation rate.apoptosis exponent and some proteins related to proliferation and apoptosis were detected. Results:Our study suggest that over-expressed of Cyr61 could enhance the expression of ECM component, promote the proliferation induced by 10ng/ml EGF.inhibit the apoptosis induced by serum-free DMEM. Cyr61 protein expression in cyst-lining epithelial cells could be reduced by RNA interfere, which could inhibit the cell proliferation and expression of ECM component,promote the apoptosis induced by serum-free DMEM.The promoting proliferation and resisting apoptosis were partly carried out through Erk and Akt pathway.
