[Background] Mechanisms of C.neoformans that cause both penetrating BBB and disseminate CNS are now under investigation. Found out that serine proteinase improves open the level of BBB, serine proteinase possess ability of philo- nerves, several authors have described that the C.neoformans secreted serine proteinases. [Objective] Mechanism of the Cryptococcal Yeast Cells Penetrating BBB into Brain Mediated by Serine Proteases and its effect to CNS. [Method] Construction of strain microarray, Immunohistochemistry, Clearance Halos, substrate gel analysis, TEM, Steedman-Lison, Fluorescence microscope, et al to make use of experiment. [Result] In vitro: Thirty-three isolates of thirty-six ones produced distinct clearance halos (CH) on the agar plates with azoalbumin; Observed positive expression of serine protease among all strains; Intervention factors effective can be diminish an area of cell becamed only 20% of original ones; In vivo: detected the expression of Azo-Blue fluorescence at BBB encephalic region; Serine protease to cause increased amounts of yeast cells burdens penetrated BBB and decurtated time of penetrated BBB. [Conclusion] All C.neoformans contain serine protease; Serine proteinase perturbs the integrity of BMEC of the blood-brain barrier, improves open the level of BBB; Secreted serine protease from pathogenic fungi were virulence.
